摘  要:目的 探讨大鼠急性心肌梗死发生后3、6 h内通过激动PINK1/Parkin通路对心肌细胞的影响。方法 将36只大鼠随机分为假手术组、模型组、PINK1激动组,每组12只;建模后按第3、6 h分为2个亚组,每组6只。模型组、PINK1激动组采用冠状动脉结扎法制备大鼠心肌梗死模型,动物模型实验结束后,摘取大鼠心脏制成切片,光镜下观察各组急性心肌梗死大鼠组织细胞的病理结构。结果 假手术组大鼠心肌细胞排列整齐,未见明显炎性细胞浸润。模型组大鼠在3、6 h时间点均观察到大量心肌细胞坏死。在3 h时间点PINK1激动组与3 h时间点模型组相比,炎性细胞减少,心肌细胞排列相对整齐,症状有所改善;6 h时间点PINK1激动组与6 h时间点模型组相比无明显差别。3 h时间点的PINK1激动组损伤程度小于同时间点模型组(P<0.05),而6 h时间点的PINK1激动组损伤程度与同时间点模型组比较无显著性差异(P>0.05)结论 大鼠发生急性心肌梗死后3 h内激动PINK1/Parkin通路可减轻大鼠心肌细胞损伤。Objective To investigate the effects of PINK1/Parkin Signaling Pathway on myocardial cell based on the rat model of acute myocardial infarction(AMI)3 and 6 hours after the cardiac event.Methods 36 SPF adult male SD rats were split into four groups at random including sham operation group,AMI group and AMI+PINK1 agonist group with 12 rats in each group.Each group was divided into two subgroups of 3 hours and 6 hours,with 6 rats in each subgroup.The rat model of AMI in AMI group and AMI+PINK1 agonist group was ligated with left anterior descending coronary artery.The myocardial cells were observed using a light microscope after the surgery.Results HE staining results showed that among the sham groups the myocardial cells of rats were ranked regularly and no inflammatory cell infiltration was observed,but the myocardial cells showed massive fused necrosis in the AMI group at 3 h and 6 h time points.Compared to the AMI groups,the inflammatory cells in the AMI+PINK1 agonist groups at 3 h time points were considerably reduced,and the myocardial cells were ranked better than it.The results of the AMI+PINK1 agonist groups at 6 h time points were similar to the AMI groups.Damage degree was less severe in the AMI+PINK1 agonist groups at 3 h time points compared to the AMI groups(P<0.05).While there was no significant difference in the degree of myocardial injury between AMI group and AMI+PINK1 agonist groups at 6 h time points(P>0.05).Conclusion Activation of PINK1/Parkin pathway within 3 hours after AMI in rats can reduce myocardial cell injury in rats.
