摘 要:微生物转化法制备雄甾-4-烯-3,17-二酮(4-AD)羟化物,具有成本低、污染小、安全、高效、专一性强等优点。本文以4-AD为底物,筛选的出发菌为泡盛曲霉MH-18菌株,底物最佳投入时间是48h,目标产物是11α-羟基-雄甾-4-烯-3,17-二酮(11α-OH-4-AD),最佳培养条件(mg/mL)是糊精33.6、玉米粉10.5、磷酸二氢钾1.2、pH4.5,最佳转化条件是温度32℃、摇床转速200r/min、装液量100mL发酵培养基/500mL摇瓶装、助溶剂甲基-β-环糊精、底物浓度1g/L、反应时间96h。转化工艺优化后,较优化前,11α-OH-4-AD产率提高35.52%,比现有的文献报道提高7.54%,具有极好的工业化应用前景。The preparation of androsterol-4-ene-3,17-dione(4-AD)hydroxylate by microbial transformation has the advantages of low cost,low pollution,safety,high efficiency and strong specificity.In this paper,4-AD is used as the substrate,the starting strain screened was Aspergillus fumigatus mh-18,the optimal input time of substrate is 48h,target products are 11α-Hydroxy androsterol-4-ene-3,17-dione(11α-OH-4-AD),the optimum culture conditions(mg/ml)are dextrin 33.6,corn flour 10.5,potassium dihydrogen phosphate 1.2 and pH4.5,and the optimal transformation conditions are temperature 32℃,rotating speed of shaking table 200r/min,volume of 100ml fermentation medium/500ml shaking flask,cosolvent methyl-β-Cyclodextrin,substrate concentration 1g/L,reaction time 96h.After optimization of the conversion process,compared with before optimization,the yield of 11α-OH-4-AD is increased by 35.52%,7.54%higher than that reported in the existing literature,and it has an excellent prospect of industrial application.
