摘 要:为探明大王杜鹃(Rhododendron rex)根际土壤细菌群落结构,为今后开展大王杜鹃的根际促生菌(PGPR)分离筛选、人工培育及濒危机制研究奠定基础,本研究以野生大王杜鹃根际土壤为研究对象,采用高通量测序技术对土壤细菌16S rRNA的V3-V4区片段进行测序。结果表明:大王杜鹃根际土壤细菌测序共获得有效序列230150条,基于≥97%的相似阈值对有效序列进行聚类后获得958个操作分类单元(OTU),隶属于19门54纲83目140科188属。细菌群落呈多样性分布,且在门、纲、目、科、属分类水平上的优势类群较为突出,优势菌门为变形菌门(Proteobacteria)、酸杆菌门(Acidobacteria)、放线菌门(Actinobacteria)、浮霉菌门(Planctomycetes)、绿弯菌门(Chloroflexi)和疣微菌门(Verrucomicrobia)。细菌生态功能以化能异养(Chemoheterotrophy)、需氧化能异养(Aerobic chemoheterotrophy)和固氮作用(Nitrogen fixation)为主,种群的相对丰度分别为6.98%、6.65%和4.31%。In this study,the rhizosphere soil of wild Rhododendron rex was taken as the research object and the V3-V4 fragment of 16S rRNA of soil bacteria was sequenced using high-throughput sequencing tech-nology to explore the bacterial community structure of rhizosphere soil,and lay foundations for isolation and screening,artificial cultivation of plant growth promoting rhizobacteria(PGPR)and endangered mechanism study.The results showed that a total of 230150 valid sequences were obtained,and 958 OTUs were obtained based on the similarity threshold of≥97%,belonged to 19 phyla,54 classes,83 orders,140 families,188 genera.The bacterial community was distributed in diversity,and the dominant groups were prominent at the classification level of phylum,class,order,family and genus.Proteobacteria,Acidobacteria,Actinobacteria,Planctomycetes,Chloroflexi and Verrucomicrobia were the dominant phylum.The main ecological functions of bacteria were chemoheterotrophy,aerobic chemoheterotrophy and nitrogen fixation,with the relative abundance as 6.98%,6.65%and 4.31%,respectively.
