摘 要:通过去卵巢大鼠骨质疏松模型,考察壮骨伸筋胶囊(ZSC)干预给药对骨质疏松大鼠的作用及其机制。实验中将造模后大鼠随机分为模型组、结合雌激素组(0.1 mg/kg)和ZSC组(2.71 g/kg),并设置假手术组,每组6只。干预给药13周后,取血清检测超氧化物歧化酶(SOD)活性和丙二醛(MDA)、钙(Ca)、磷(P)、白介素-1β(IL-1β)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)的含量;解剖检测各组大鼠股骨长度、质量、密度、病理变化,Western Blot检测ZSC对大鼠股骨组织中血红素加氧酶-1(HO-1)、核因子E2相关因子2(Nrf-2)和核因子κB(NFκB)蛋白表达的影响。实验结果表明,与模型组相比,ZSC大鼠股骨质量升高(P<0.01)、股骨密度升高(P<0.05);与模型组相比,ZSC大鼠血清中MDA、IL-1β、IL-6、TNF-α、血P降低(P<0.01),血清中SOD水平升高(P<0.01)、血Ca含量升高(P<0.01);与模型组相比,ZSC大鼠Nrf-2、HO-1蛋白表达水平提高(P<0.01),NFκB蛋白表达降低(P<0.05)。由以上结果可知,ZSC对去卵巢导致的大鼠骨质疏松症有一定改善作用,其作用机制可能与其抗炎、抗氧化活性相关。Ovariectomized rat model of osteoporosis was used to investigate the effects of Zhuanggu Shenjin Capsule(ZSC)on osteoporosis and its mechanism.Ovariectomized rats were randomly divided into model group,combined estrogen group(0.1 mg/kg)and ZSC group(2.71 g/kg).A sham group was also used in the experiment,each group allocates 6 rats.After 13 weeks,the activity of superoxide dismutase(SOD)and the contents of malondialdehyde(MDA),calcium(Ca),phosphorus(P),interleukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)were detected in the serum.The length,weight,bone mineral density,pathology and proteins(HO-1,Nrf-2 and NFκB)expression of femur in each group were examined.Compared with the model group,the weight of femur in ZSC group significantly increased(P<0.01)and the bone mineral density also significantly increased(P<0.05).Compared with the model group,the contents of MDA,IL-1β,IL-6,TNF-αand P significantly reduced(P<0.01),while the content of Ca and the activity of SOD significantly increased(P<0.01)in ZSC group.Compared with the model group,the expression of Nrf-2 and HO-1 significantly increased(P<0.01),while the expression of NFκB(P<0.05)significantly decreased in ZSC group.ZSC has certain improvement on osteoporosis caused by ovariectomizing in rats,and its mechanism may be related to its anti-inflammatory and antioxidant activities.
