摘 要:目的探讨矮小相关转录因子1(RUNX1)在不明原因复发性流产(URSA)病例中的表达与调控作用。方法选取2022年3月至10月广西医科大学第一附属医院收治的5例URSA患者的绒毛组织作为实验组,选取同期5例行人工流产的妊娠女性的正常妊娠绒毛组织作为对照组。利用全基因组高通量测序,挖掘差异表达基因;利用生物信息学网站GeneMania预测RUNX1与血管生成的关系。结果共发现2526个差异表达mRNA,其中上调1815个,下调711个,其中实验组中的RUNX1表达上调(P<0.01)。RUNX1与血管生成信号通路相关。结论RUNX1与早期胚胎发育相关,可能通过抑制血管生成导致胚胎停育。Objective To investigate the expression and regulation role of runt-related transcription factor 1(RUNX1)in patients with unexplained recurrent spontaneous abortion(URSA).Methods The villus tissues of 5 URSA patients admitted to the First Affiliated Hospital of Guangxi Medical University from March to October 2022 were selected as the experimental group,and the normal pregnancy villus tissues of 5 pregnant women who underwent artificial abortion during the same period were selected as the control group.The whole genome high-throughput sequencing was used to explore differentially expressed genes.The bioinformatics website GeneMania was used to predict the relationship between RUNX1 and angiogenesis.Results A total of 2526 differentially expressed mRNA were found,of which 1815 were up-regulated and 711 were down-regulated.The expression of RUNX1 was up-regulated in the experimental group(P<0.01).RUNX1 was associated with signaling pathways involved in angiogenesis.Conclusion RUNX1 is associated with early embryonic development,which may lead to embryo termination by inhibiting angiogenesis.
