摘 要:目的观察疏血通注射液腹腔注射对慢性肾功能衰竭(CRF)大鼠肾损伤的改善作用,并探讨其机制。方法SPF级SD大鼠30只随机分为假手术组、模型组、疏血通注射液组,每组10只。模型组、疏血通注射液组大鼠采用分阶段5/6肾切除法制备大鼠CRF模型。CRF大鼠模型建立成功后,疏血通注射液组按10 mL/(kg•d)的剂量腹腔注射疏血通注射液,假手术组、模型组注射同剂量的生理盐水,连续8周后,收集腹主动脉血清、摘取肾脏备用。采用HE染色法观察各组大鼠肾脏组织病理学变化。使用全自动生化仪及相应试剂盒检测尿素氮(BUN)、肌酐(Scr)、晚期蛋白质氧化产物(AOPPs)、活性氧(ROS)、丙二醛(MDA)、超氧化物歧化酶(SOD)等肾功能标志物和氧化应激标志物。采用JC-1荧光探针法检测线粒体膜电位(MTP),使用ATP活性试剂盒检测ATP含量。采用Western Blotting法检测大鼠肾组织沉默信息调节因子1(SIRT1)、叉头框转录因子O1(FoxO1)蛋白。结果模型组大鼠肾小管变形、扩张、萎缩,肾间质可见较多炎症细胞浸润及上皮细胞坏死,伴有腺嘌呤沉积;疏血通给药组大鼠肾间质的炎性细胞浸润减少,肾小管状态改善,腺嘌呤结晶有所减少,病理损伤相对较轻。与假手术组相比,模型组、疏血通注射液组大鼠BUN、Scr、ROS、MDA、AOPPs水平均升高(P均<0.05),SOD活性、ATP酶含量、MTP均降低(P均<0.05);与模型组相比,疏血通注射液组大鼠BUN、Scr、ROS、MDA、AOPPs水平均降低(P均<0.05),SOD活性、ATP酶含量、MTP均升高(P均<0.05)。与假手术组相比,模型组、疏血通注射液组大鼠肾组织SIRT1蛋白相对表达量均降低(P均<0.05),模型组大鼠肾组织FoxO1蛋白相对表达量升高(P<0.05);与模型组相比,疏血通注射液组大鼠肾组织SIRT1蛋白相对表达量升高(P<0.05),FoxO1蛋白相对表达量降低(P<0.05)。结论疏血通注射液腹腔注射对CRF大鼠的肾损伤具有改善作用,�Objective To investigate the effect of intraperitoneal injection of the Shuxuetong injection on kidney injury rats with chronic renal failure(CRF),and to explore its mechanism.Methods Thirty SPF-grade SD rats were random divided into sham operation group,model group,and Shuxuetong injection group,with 10 rats in each group.In the model group and the Shuxuetong injection group,the rat CRF models were constructed by staged 5/6 nephrectomy.After the rat CRF models were successfully constructed,the Shuxuetong injection group was injected intraperitoneally at a dose of 10 mL/(kg•d),while the sham group and the model group were injected with the same dose of normal saline.After 8 consecutive weeks,abdominal aorta serum was collected and kidneys were collected for backup.HE staining was used to observe the histopathological changes of rat kidneys in each group.Automatic biochemical analyzer and corresponding kit were used to detect renal function markers and oxidative stress markers such as urea nitrogen(BUN),creatinine(Scr),advanced oxidative protein products(AOPPs),reactive oxygen species(ROS),malondialdehyde(MDA),and superoxide dismutase(SOD).The mitochondrial membrane potential(MTP)was detected by JC-1 fluorescent probe method,and the ATP content was detected by using the ATP activity kit.Western blotting was used to detect rat kidney tissue silencing information regulator 1(SIRT1)and forkhead box transcription factor O1(FoxO1).Results In the model group,renal tubular deformation,dilation and atrophy were observed,and more inflammatory cell infiltration and epithelial cell necrosis were observed in the renal interstitium,accompanied by adenine deposition.In the Shuxuetong injection group,the inflammatory cell infiltration in the renal interstitium was reduced,the tubular condition was improved,adenine crystals were reduced,and the pathological damage was relatively mild.Compared with the sham operation group,the levels of BUN,SCR,ROS,MDA and AOPPs in the model group and the Shuxuetong injection group increased(all
