摘  要:目的:探讨笃斯越橘对氧化损伤心肌细胞的作用。方法:以笃斯越橘果实为原料提取得到粗提物1个,并进一步纯化得到4个柱层析组分,建立H_(2)O_(2)氧化模型,考察提取物及纯化产物对氧化损伤细胞存活率的影响,并对最优组分进行成分分析。结果:笃斯越橘粗提物及4种纯化产物均能提高氧化损伤H9c2心肌细胞的存活率,其中30%乙醇水溶液洗脱组分效果最好,能有效减少ROS和MDA生成,提高SOD活性;对该组分进行超高效液相色谱-质谱联用(UPLC-MS)分析,鉴定出花色苷14种。结论:笃斯越橘果实提取及纯化产物均对H_(2)O_(2)引起的H9c2心肌细胞氧化损伤具有保护作用。Objective To investigate the effect of Vaccinium uliginosum on oxidative damage of cardiomyocytes.Method One crude extract was extracted from the fruit and further four column chromatographic components were obtained.An H_(2)O_(2) oxidation model was established to investigate the effects of the extract and purified products on the survival rate of oxidative damage cells,and the optimal components were analyzed.Result The extract of Vaccinium uliginosum fruit and four column chromatography purified products can improve the survival rate of H9c2 cardiomyocytes.Among them,30%ethanol aqueous solution has the best effect,which can effectively reduce the production of ROS and MDA and improve the activity of SOD.By UPLC-MS analysis,14 kinds of anthocyanins were identified.Conclusion The extract of Vaccinium uliginosum fruit and four column chromatography purified products have protective effects on the oxidative damage of H9c2 cardiomyocytes caused by H_(2)O_(2).
