摘 要:目的基于miR-126a-3p调控探讨皮肤原位再生医疗技术(moist exposed burn therapy/moist exposed burn ointment,MEBT/MEBO)对糖尿病大鼠慢性创面血管新生的影响。方法72只SPF级Wistar雄性大鼠,随机均分为对照组、模型组及MEBO组。构建急性创面和糖尿病慢性创面模型,对照组和模型组均用生理盐水处理,MEBO组用MEBO处理。对比各组创面愈合率,形态学变化,毛细血管数及miR-126a-3p表达量。结果治疗后第11天及第18天,各组创面愈合率从大到小排列为对照组、MEBO组、模型组,并且,治疗后第18天各组大鼠创面愈合率均高于第11天(P均<0.001)。治疗后第18天模型组和MEBO组毛细血管数量相对于治疗后第1天均明显增加(P<0.001),但MEBO组毛细血管数量增加比模型组更明显(P<0.001)。治疗后第18天,与模型组相比,MEBO组创面组织中miR-126a-3p表达量显著增加(P<0.001)。结论MEBT/MEBO可能通过上调miR-126a-3p促血管新生,从而促进糖尿病慢性创面愈合。Objective To investigate the effect of moist exposed burn therapy/moist exposed burn ointment(MEBT/MEBO)on angiogenesis in chronic wound tissue of diabetic rats based on the regulation of miR-126a-3p.Methods 72 SPF grade Wistar male rats were randomly divided into control group,model group,and MEBO group.Acute wound and diabetic chronic wound models were constructed,and both the control group and the model group were dealed with normal saline,and the MEBO group were dealed with MEBO.And then,wound healing rates,morphological changes,capillary number,and miR-126a-3p expression were compared between groups.Results On the 11 th and 18 th day after treatment,the wound healing rates ranked from large to small were the control group,the MEBO group,and the model group.Moreover,the wound healing rates in each group on the 18 th day after treatment were higher than those on the 11 th day after treatment(all P<0.001).The number of capillaries in the model group and the MEBO group increased significantly on the 18 th day after treatment compared to the first day after treatment(P<0.001),but the increase in the number of capillaries in the MEBO group was more significant than that in the model group(P<0.001).On the 18 th day after treatment,compared with the model group,the expression of miR-126a-3p in the wound tissue of the MEBO group significantly increased(P<0.001).Conclusion MEBT/MEBO may promote angiogenesis by up regulating miR-126a-3p,and thus promoting chronic wound healing in diabetes.
