活血通络汤对抗中性粒细胞胞浆抗体相关性血管炎患者内皮祖细胞功能及血管内皮损伤因子的影响

摘　　要：目的 观察活血通络汤对抗中性粒细胞胞浆抗体相关性血管炎(AAV)患者外周血内皮祖细胞(EPCs)功能及血管内皮损伤因子的影响,探讨其作用机制。方法 采用密度梯度离心法分离AAV患者外周血EPCs进行培养,将EPCs分为正常组、模型组、空白血清组(10%空白血清)和活血通络汤低(5%含药血清)、中(10%含药血清)、高(20%含药血清)剂量组,干预48 h,MTT法检测细胞增殖能力,Transwell小室检测细胞迁移能力,检测细胞黏附能力,Western blot检测EPCs血管性假性血友病因子(vWF)、可溶性血栓调节蛋白(sTM)表达。结果 与正常组比较,模型组EPCs增殖、迁移、黏附能力显著降低(P<0.01),vWF、sTM蛋白表达显著升高(P<0.01);与模型组及空白血清组比较,活血通络汤各剂量组EPCs增殖、迁移及黏附能力显著升高(P<0.05,P<0.01),vWF、sTM蛋白表达显著降低(P<0.05,P<0.01),以活血通络汤高剂量组作用最显著(P<0.01)。结论 活血通络汤可能通过提高EPCs增殖、迁移及黏附能力,降低血管内皮损伤因子vWF、sTM蛋白表达,从而治疗AAV。Objective To observe the effects of Huoxue Tongluo Decoction on endothelial progenitor cells(EPCs) of function and vascular endothelial injury factors in patients with ANCA associated vasculitis(AAV);To discuss its mechanism of action.Methods Peripheral blood EPCs were separated by density gradient centrifugation for culture.EPCs were divided into normal group,model group,blank serum group(10% blank serum) and Huoxue Tongluo Decoction low-(5% medicated serum),medium-(10% medicated serum) and high-(20% medicated serum)dosage groups.After 48 hours of intervention,the cell proliferation was detected by MTT method,Transwell chamber was used to detect the cell migration ability,cell adhesion was detected,Western blot was used to detect the expressions of von Willebrand factor(vWF) and soluble thrombomodulin(sTM) in EPCs.Results Compared with the normal group,the proliferation,migration ability and adhesion ability of EPCs in the model group decreased significantly(P<0.01),and the expressions of vWF and sTM protein significantly increased(P<0.01).Compared with the model group and blank serum group,the proliferation,migration and adhesion ability of EPCs significantly increased in all dosage of Huoxue Tongluo Decoction groups(P<0.05,P<0.01),the expressions of vWF and sTM protein significantly decreased(P<0.05,P<0.01),and the Huoxue Tongluo Decoction high-dosage group showed the most obvious effect(P<0.01).Conclusion Huoxue Tongluo Decoction may treat AAV by improving the proliferation,migration and adhesion ability of EPCs and reducing the expressions of vascular endothelial injury factors vWF and sTM protein.
