摘 要:为探究淫羊藿素和山柰酚对鸡卵泡发育的作用,试验通过MTT法分析不同浓度的淫羊藿素和山柰酚对鸡等级前颗粒细胞活性的影响并确定最佳浓度,通过实时荧光定量PCR检测淫羊藿素和山柰酚对鸡等级前颗粒细胞增殖细胞核抗原(PCNA)、促卵泡素受体(FSHR)和雌激素受体α(ERα)mRNA表达的影响;通过卵巢组织离体培养,分析淫羊藿素和山柰酚对卵巢组织卵泡发育的影响。结果显示:淫羊藿素和山柰酚均在浓度为1×10^(-4)mol/L时可以显著促进颗粒细胞的增殖,同时可以促进颗粒细胞中PCNA、FSHR和ERαmRNA的表达(P<0.01);淫羊藿素和山柰酚作用的卵巢组织中的卵泡数量明显增多,且颗粒细胞由单层增殖至多层。研究表明,淫羊藿素和山柰酚能够促进等级前卵泡颗粒细胞的增殖,促进FSHR和ERαmRNA的表达,从而促进等级前卵泡向等级卵泡的发育。To reveal the effects of icaritin and kaempferol on proliferation of prehierarchical and preovulatory follicles cells in chicken,the effects of different concentrations of icaritin and kaempferol on the activity of chicken prehierarchical and preovu⁃latory follicles cells were analyzed by MTT assay and the optimal concentration was determined,then the effects of icaritin and kaempferol on the expression of proliferatingcell nuclear antigen(PCNA),follicle-stimulating hormone receptor(FSHR)and es⁃trogen receptorα(ERα)mRNA in chicken prehierarchical and preovulatory follicles cells were detected by real-time PCR;The effects of icaritin and kaempferol on follicular development in ovarian tissues were analyzed by ovarian tissue culture.The results showed that both icaritin and kaempferol could significantly promote the proliferation of granulosa cells at a concentration of 1×10^(-4)mol/L,and at the same time,they could promote the expression of PCNA,FSHR and ERαmRNA in prehierarchical and preovulatory follicles cells(P<0.01).The number of follicles in ovarian tissue treated with icaritin and kaempferol was significant⁃ly increased,and the granulosa cells developed from a monolayer to multiple layers.The results indicated that icaritin and kaemp⁃ferol could promote the proliferation of granulosa cells of prehierarchical and preovulatory follicles and promote the expression of FSHR and ERαmRNA,then promoting the development of prehierarchical and preovulatory follicles.
