不同浓度新鲜蛋清对大鼠足跖肿胀急性炎症模型的研究

摘　　要：目的通过对不同浓度新鲜蛋清所致的大鼠足跖肿胀研究,优选出新鲜蛋清所致的大鼠足跖肿胀急性炎症模型条件。方法选用蛋清原液、50%蛋清和10%蛋清3个浓度,于大鼠后足足跖部位皮下注射,分别检测致炎前和致炎后0.5、1、2、3和4 h大鼠的足跖肿胀度,优选出蛋清所致的大鼠足跖肿胀急性炎症模型最佳造模浓度和作用时间,并采用阳性药阿司匹林对优选建立的大鼠足跖肿胀模型条件进行验证。结果与10%蛋清相比,50%蛋清和蛋清原液在致炎后1、2和4 h,大鼠的足跖肿胀度升高(P<0.05);足跖肿胀度在1 h达到高峰,蛋清原液、50%蛋清、10%蛋清分别为(0.80±0.08、0.78±0.09、0.40±0.13)ml,而后逐渐下降,但二者致炎效果无明显差别(P>0.05)。以50%蛋清浓度作用1 h为造模条件,结果显示阿司匹林组的肿胀度和TNF-α含量低于50%蛋清组(P<0.05)。结论50%蛋液浓度作用1 h为最佳的大鼠足跖肿胀急性炎症模型条件。Objective To investigate the optimal conditions of rat paw oedema model induced by implying egg albumen on foot swelling of rats with different concentrations.Methods Three concentrations of egg albumen(egg albumen stock solution,50%egg albumen and 10% egg albumen)were selected and subcutaneously injected into the hind paw of rats.The paw swelling degree of rats before inflammation and 0.5,1,2,3 and 4 h after inflammation was detected respectively,and the optimal concentration and action time of acute inflammation model of rat paw swelling induced by egg white were selected.Aspirin was used to verify the optimal conditions of the rat paw oedema model.Results Compared with the 10% egg albumen,the degree of foot swelling of the rats was significantly increased at 1,2 and 4 h after 50% and 100% egg albumen were used(P<0.05).The degree of plantar swelling reached its peak at 1 h,the egg albumen stock solution,50%egg albumen and 10% egg albumen were(0.80±0.08)ml,(0.78±0.09)ml and(0.40±0.13)ml,respectively,and then gradually decreased,but there was no significant difference in the inflammatory effect between the two groups(P>0.05).50%egg albumen concentration for 1 h as the modeling condition,the results showed that the swelling degree and TNF-α content in aspirin group were lower than those in 50% egg white albumen(P<0.05).Conclusion The optimum condition for the rat model of foot swelling is 50% egg solution for 1 h.
