摘　　要：目的 通过生物信息学方法研究胶原三股螺旋重复蛋白1(CTHRC1)基因在胃癌组织与正常组织中的表达差异,并分析其与胃癌患者预后和免疫细胞浸润水平的关系。方法 通过GEPIA、UALCAN数据库探讨CTHRC1基因在胃癌组织中的表达水平及其与胃癌患者病理分期的关系;Kaplan-Meier Plotter数据库分析CTHRC1基因表达水平与胃癌患者预后的关系;STRING数据库分析CTHRC1相关蛋白网络并用Metascape数据库进行基因本体(GO)和京都基因与基因组百科全书(KEGG)富集分析;TIMER数据库分析CTHRC1基因表达与胃癌微环境中不同类型免疫细胞的相关性。结果 CTHRC1基因在多种癌症中呈现高表达,在胃癌组织中的表达水平显著高于正常组织,与胃癌病理分期密切相关(P <0.05);CTHRC1基因高表达与胃癌患者不良预后成显著相关性(P <0.05)。蛋白质互作网络分析揭示了CTHRC1与DVL1、DVL2、DVL3、FZD3、FZD5、FZD6、ROR2等蛋白具有明显相互作用,富集分析结果显示上述蛋白与Wnt/PCP信号通路相关。CTHRC1基因表达与胃癌微环境中B细胞的浸润水平成负相关,与CD8^(+)T细胞、巨噬细胞、中性粒细胞、树突状细胞的浸润水平成正相关(P <0.05)。结论 CTHRC1基因在胃癌组织中高表达,与胃癌的发生、发展及预后密切相关,其影响胃癌发生、发展的分子机制可能与调节Wnt/PCP信号通路及影响免疫细胞浸润水平相关。Objective To analyze the differential expression of collagen triple helix repeat containing 1(CTHRC1)gene in gastric cancer tissues and normal tissues by bioinformatics analysis,and to determine its relationship with gastric cancer prognosis and immune cells infiltration level.Methods GEPIA and UALCAN databases were used to determine the expression level of CTHRC1 gene in gastric cancer tissues and its relationship with pathological stage of patients with gastric cancer.Kaplan-Meier Plotter database was used to analyze the relationship between CTHRC1 gene expression level and prognosis of patients with gastric cancer.The CTHRC1-related protein networks were analyzed by STRING database.Gene Ontology(GO),and Kyoto Encyclopedia of Genes and Genomes(KEGG)were enriched by Metascape database.TIMER database was used to analyze the correlation between CTHRC1 gene expression and different types of immune cells in gastric cancer microenvironment.Results CTHRC1 gene was highly expressed in most cancers,and its expression level in gastric cancer tissues was much higher than that in normal tissues, which was closely related to pathological stage of gastric cancer (P < 0.05). High expression of CTHRC1 gene was also significantly correlated with the poor prognosis of patients with gastric cancer (P < 0.05). Protein interaction network analysis revealed that CTHRC1 had significant interaction with DVL1, DVL2, DVL3, FZD3, FZD5, FZD6, ROR2 and other proteins. GO and KEGG enrichment analysis were performed for these proteins. The expression of CTHRC1 gene was negatively correlated with the infiltration level of B cells in gastric cancer microenvironment, but positively correlated with the infiltration level of CD8 ^(+) T cells, macrophages, neutrophils and dendritic cells (P < 0.05). Conclusion High expression of CTHRC1 gene in gastric cancer tissues is closely related to its occurrence, development and prognosis.
