摘 要:目的筛选瘀血痹胶囊中保护心肌细胞损伤的药效物质。方法采用HPLC法建立瘀血痹胶囊7个不同极性部位指纹图谱,共获得41个共有峰;以阿霉素建立H9c2心肌细胞损伤的体外药效模型,将心肌细胞存活率作为药效指标,开展不同极性的瘀血痹胶囊体外药效学评价;应用灰色关联度分析结合偏最小二乘回归分析,将指纹图谱共有峰相对峰面积与保护心肌细胞药效作用进行关联,筛选瘀血痹胶囊中保护心肌细胞损伤的药效物质;并对药效物质的心肌保护作用进行验证。结果筛选出瘀血痹胶囊保护心肌细胞损伤的5个药效物质,其中指认出的主要药效物质为槲皮素、隐丹参酮、丹参酮ⅡA和阿魏酸。结论瘀血痹胶囊具有心肌细胞损伤保护作用,通过建立H9c2心肌细胞损伤模型进行药效物质筛选,初步明确了瘀血痹胶囊保护心肌细胞损伤的主要药效物质。Objective To determine the protective effect of pharmacodynamic substances in Yuxuebi capsules on myocardial cell damage.Methods The HPLC fingerprints of the 7 polar parts of Yuxuebi capsules were established,and 41 common peaks were obtained.In vitro pharmacodynamic model of H9c2 myocardial cell damage was established with adriamycin,and the vitro pharmacodynamic evaluation of Yuxuebi capsules of different polarities was conducted with the survival rate of cardiomyocytes as a pharmacodynamic index.Grey correlation analysis combined with mathematical methods of partial least squares regression analysis were used.The relative peak area of the total peak of fingerprints correlated with the pharmacodynamic effect of protecting cardiomyocytes,and the pharmacodynamic substances in Yuxuebi capsules to protect the myocardial cell damage were screened.The myocardial protective effect of the medicinal agent was verified.Results Five pharmacodynamic substances were screened out with protection against myocardial cell damage in Yuxuebi capsules, the main pharmacoactive substances being quercetin, cryptotanshinone, tanshinone ⅡA and ferulic acid. Conclusion Yuxuebi capsules can protect against myocardial cell damage, and the main pharmacodynamic substances are preliminarily established by H9c2 myocardial cell damage model.
