摘 要:目的:探究在慢性阻塞性肺疾病的炎症环境中,清金化痰汤通过调节炎症因子在调控铁代谢发挥治疗作用的机制研究。方法:选用气管滴入脂多糖(LPS)结合烟熏的方法制备COPD模型。随机将40只大鼠分成正常组、COPD组、罗红霉素西药组,清金化痰汤中药组(n=10)。造模后给药干预,清金化痰汤中药组灌服14.62 g/kg(清金化痰汤),罗红霉素西药组灌服0.01575 g/kg(罗红霉素),COPD组及正常组以生理盐水10 mL/kg,一日2次。ELISA法检测每组大鼠血清中铁调素(Hepcidin)、IL⁃1β、IL⁃10、TGF⁃β1的表达;Western Blot及qRT⁃PCR检测每组大鼠肺组织中TF、IREB2 mRNA及蛋白的表达。结果:较正常组,COPD组血清Hepcidin、IL⁃10的含量明显降低(P<0.01),血清IL⁃1β、TGF⁃β1的含量明显升高(P<0.01,P<0.05)。较COPD组,两药物组血清Hepcidin、IL⁃10的含量明显升高(P<0.01),血清IL⁃1β、TGF⁃β1的含量明显下降(P<0.05);较正常组,COPD组肺组织中TF mRNA及蛋白表达升高(P<0.01),IREB2 mRNA及蛋白表达下降(P<0.01);较COPD组,两用药组肺组织中TF mRNA及蛋白表达下降(P<0.01,P<0.05),IREB2 mRNA及蛋白表达上升(P<0.01)。结论:铁代谢与炎症相关,清金化痰汤可以在炎症环境调节铁代谢治疗COPD。Objective:To explore the mechanism of Qingjin Huatan Decoction in regulating iron metabolism by regulating inflammatory factors in the inflammatory environment of chronic obstructive pulmonary disease.Methods:The COPD model was prepared by tracheal drops of lipopolysaccharide(LPS)combined with smoke.40 rats were randomly divided into normal group,COPD group,roxithromycin western medicine group and Qingjin Huatan Decoction traditional Chinese medicine group(n=10).Post⁃molding drug administration intervention,Qingjin Huatan Decoction Chinese medicine group was given 14.62 g/kg(Qingjin Huatan Decoction),roxithromycin western medicine group was given 0.01575 g/kg(roxithromycin),COPD group and normal group were given normal saline 10 mL/kg,twice a day.The expressions of Hepcidin,IL⁃1β,IL⁃10 and TGF⁃β1 in serum of rats in each group were detected by ELISA.WesternBlot and qRT⁃PCR were used to detect the expression of TF and IREB2 protein and mRNA in lung tissues of each group.Results:Compared with the normal group,the contents of serum Hepcidin and IL⁃10 in COPD group were significantly decreased(P<0.01),while the contents of serum IL⁃1βand TGF⁃β1 were significantly increased(P<0.01,P<0.05).Compared with COPD group,the contents of serum Hepcidin and IL⁃10 were significantly increased in the two drug groups(P<0.01),while the contents of serum IL⁃1βand TGF⁃β1 were significantly decreased in the two drug groups(P<0.05).Compared with normal group,TF mRNA and protein expression in COPD group were increased(P<0.01),while IREB2 mRNA and protein expression were decreased(P<0.01).Compared with COPD group,TF mRNA and protein expres⁃sions in lung tissues of the two⁃drug group were decreased(P<0.01,P<0.05),while IREB2 mRNA and protein expressions were increased(P<0.01).Conclusion:Iron metabolism is related to inflammation.Qingjin Huatan Decoction can regulate iron metabolism in inflammatory environment to treat COPD.
