摘 要:目的观察启明丸对形觉剥夺性近视豚鼠干预作用及对巩膜基质金属蛋白酶-2(MMP-2)及其组织抑制物(TIMP-2)表达的影响,探讨其可能的作用机制。方法将40只2~3周龄雄性三色豚鼠,随机分为高剂量启明丸组(HQMG)、低剂量启明丸组(LQMG)、模型组(MG)及空白组(BG)4组,每组10只。用形觉剥夺法建立豚鼠的近视模型,4周确认造模成功后,HQMG和LQMG豚鼠分别给予启明丸药粉1.44 g/200 g、0.36 g/200 g溶于0.9%氯化钠注射液2 mL灌胃;MG与BG豚鼠给予0.9%氯化钠注射液2 mL灌胃。各组均每日1次,共4周。观察各组豚鼠屈光度、眼轴长度变化,巩膜组织病理学改变,并采用Western Blot检测巩膜组织中MMP-2、TIMP-2的表达。结果(1)近视模型:造模4周后,各造模组实验眼近视屈光度与自身对照眼相比均增加(t_(HQMG)=-5.204,t_(LQMG)=-5.626,t_(MG)=-4.283,均P=0.000);4组间实验眼近视屈光度变化两两比较,BG均低于各造模组,差异均有统计学意义(t_(HQMG)=-3.690,t_(LQMG)=-3.850,t_(MG)=-3.969,均P=0.001)。各造模组实验眼与自身对照眼相比眼轴长度明显增长(t_(HQMG)=2.440,P=0.025;t_(LQMG)=2.141,P=0.045;t_(MG)=2.341,P=0.025)。4组间实验眼眼轴长度变化两两比较,BG均低于各造模组(t_(HQMG)=3.881,P=0.001,t_(LQMG)=3.115,P=0.006,t_(MG)=2.600,P=0.019),差异均有统计学意义。(2)屈光度变化:造模8周后,3组间实验眼近视屈光度两两比较,MG较各治疗组增加(t_(HQMG)=2.883,P=0.010;t_(LQMG)=2.442,P=0.030),差异均有统计学意义。灌胃前后3组间近视屈光度差值两两比较,MG高于HQMG,差异有统计学意义(t=2.999,P=0.008)。(3)眼轴长度变化:3组间实验眼眼轴长度两两比较,MG较各治疗组增加(t_(HQMG)=-2.422,P=0.026;t_(LQMG)=-2.372,P=0.029),差异均有统计学意义。灌胃前后3组间眼轴增长幅度两两比较,MG高于各治疗组(t_(HQMG)=-3.029,P=0.007;t_(LQMG)=-2.846,P=0.011),差异均有统计学意义。(4)组织病理变化:HQMG、LQMG豚鼠巩膜各层�OBJECTIVE To observe the effect of Qiming Pill on form deprivation myopia in guinea pigs and its effect on the expression of MMP-2 and TIMP-2 in sclera,and to explore its potential mechanism.METHODS Forty male guinea pigs aged 2~3 weeks were randomly divided into high-dose Qiming Pill group(HQMG),low-dose Qiming Pill group(LQMG),model group(MG)and blank group(BG),with 10 guinea pigs in each group.The myopia model of guinea pigs was established by form deprivation method.After 4 weeks,HQMG and LQMG were given 1.44 g and 0.36 g/200 g of Qiming Pill powder dissolved in 2 mL of 0.9%sodium chloride injection by gavage,respectively,while MG and BG were given 0.9%sodium chloride injection 2 mL by gavage,respectively.Each group was treated once a day for four weeks.The changes of diopter,axial length,histopathological changes of sclera were observed,and the expressions of MMP-2 and TIMP-2 in sclera were detected by Western Blot.RESULTS(1)Myopia models:After 4 weeks of modeling,the myopia diopter of experimental eyes in each model group increased compared with their own control eyes(t_(HQMG)=-5.204,t_(LQMG)=-5.626,t_(MG)=-4.283,all P=0.000).Comparing the myopia diopter of the experimental eyes between the four groups,BG was lower than that in each model group,and the difference was statistically significant(t_(HQMG)=-3.690,t_(LQMG)=-3.850,t_(MG)=-3.969,all P=0.001).Compared with the self-control eyes,the axial length of the experimental eyes in each model group increased significantly(t_(HQMG)=2.440,P=0.025;t_(LQMG)=2.141,P=0.045;t_(MG)=2.341,P=0.025).Comparing the axial length of the experimental eyes between the four groups,BG was lower than that in each model group,and the difference was statistically significant(t_(HQMG)=3.881,P=0.001;t_(LQMG)=3.115,P=0.006;t_(MG)=2.600,P=0.019).(2)Diopter changes:After eight weeks of modeling,when comparing the myopia diopter of the experimental eyes between the three groups,MG was higher than that in each treatment group,and the difference was statistically significant(t_(HQMG)=2.88
