摘 要:比较了体腔细胞与血窦游离细胞的细胞毒活性,应用转录组分析和逆转录定量PCR技术比较了多种免疫相关基因的mRNA表达水平,通过蛋白免疫印迹技术分析了AjAIF-1蛋白水平表达的差异,应用EdU体外标记DNA复制能力分析比较了两种细胞体外增殖能力的差异。结果表明,血窦游离细胞的细胞毒活性显著低于体腔细胞,免疫相关基因AjAIF-1、AjTLR3、AjToll、AjLITAF、AjC3和AjC3-2的mRNA表达水平均低于体腔细胞。AjAIF-1蛋白表达水平也明显低于体腔细胞。体外增殖能力结果显示,血窦游离细胞体外增殖能力显著高于体腔细胞。形态学观察未检测到两种细胞主要细胞类型的差异,但血窦游离细胞中振动细胞的比例显著高于体腔细胞。研究结果提示,与体腔细胞相比,血窦游离细胞可能具有较弱的免疫能力和较强的细胞增殖能力。The cytotoxic activity of coelomic cells and free blood sinus cells were compared.The mRNA expression levels of various immune related genes were compared by transcriptome analysis and reverse transcriptional quantitative PCR.The differences in the expression level of AjAIF-1 protein was analyzed by Western Blotting.The differences in the proliferation ability of two kinds of cells in vitro were compared by EdU labeled DNA replication ability analysis.The results showed that the cytotoxic activity of the isolated blood sinus cells was significantly lower than that of the coelomic cells,and the mRNA expression levels of immune related genes AjAIF-1,AjTLR3,AjToll,AjLITAF,AjC3 and AjC3-2 were lower than those of coelomic cells.The expression level of AjAIF-1 protein was also significantly lower than that of coelomic cells.The results of proliferation ability in vitro showed that the proliferation ability of isolated blood sinus cells was significantly higher than that of coelomic cells.The result of morphological observation did not showed differences in the main cell types of the two types of cells,but the proportion of vibrating cells in isolated blood sinus cells was significantly higher than that in coelomic cells.The results indicated that the isolated cells from the blood sinuses may have weaker immune abilities and stronger cell proliferation abilities compared with coelomic cells.
