摘 要:本研究旨在培育转基因耐盐碱水稻品种。利用根瘤农杆菌介导法将水稻Leap启动子调控的SNAC1基因和筛选基因Bar导入早粳稻空育131,通过分子选择、耐冷性鉴定、盐碱地原位耐盐碱性及产量鉴定来选择耐盐碱水稻。Ti质粒介导外源基因SNAC1及Bar转化水稻空育131成熟胚愈伤组织获得了55个独立株系,经多代自交鉴定选择创制了外源基因单位点整合及稳定遗传、耐冷性强的18个株系。转基因水稻株系在哈尔滨市中重度盐碱地上均具有较强的田间耐盐碱性,选择其中2个耐盐碱最强的株系为耐盐碱候选品种。在哈尔滨市中重度盐碱地逆境胁迫条件下,转基因耐盐碱候选品种呈现SNAC1基因高表达,产量性状及产量明显地优于受体水稻。转SNAC1基因耐盐碱水稻品种培育成功,在松嫩平原西部盐碱地区域具有产业化潜力。This study aimed to breed transgenic saline-alkali-tolerant rice varieties.The SNAC1 gene regu lated by leap promoter and bar gene were introduced into early japonica rice Kongyu131 by agrobacterium tumefaciens mediated method. The saline-alkali-tolerant rice was selected by molecular selection, cold tolerance identification, in-situ identification of saline alkaline tolerance and yield in saline-alkali soil.Fifty-five independent lines were obtained by using Ti plasmidto transfers SNAC1 and bargene into the mature embryo callus of rice Kongyu131.Then, eighteen lines with single locus integration of exogenous genes and stable inheritance, strong cold tolerance were obtained through identification and selection of several generation self-crossing. Under the field condition, the transgenic rice lines showed strong salinity and alkali resistance on moderate-severe saline-alkali soils in Harbin, and two of the lines with the strongest salinity and alkali tolerance were selected as candidate varieties of salinity and alkali resistance.Under the moderate-severe saline-alkali stress, these two candidate cultivars showed high expression of SNAC1 gene, and yield traits and yield were significantly better than the recipient rice.The SNAC1 transgenic saline-alkali-tolerant rice varieties were successfully bred, which has the industrialization potential in the saline-alkali soil region of western parts of Songnen Plain.
