摘 要:试验旨在利用大孔树脂纯化九节中的粗黄酮,优选纯化总黄酮的最佳工艺。试验比较3种不同大孔吸附树脂AB-8、HP-20、D-101的吸附-解吸附特性,选择最优吸附纯化树脂考察其上样工艺及洗脱工艺,采用Box-Behnken响应面分析法建立数学模型,优选最佳纯化条件。结果显示,HP-20型大孔树脂吸附性强、解吸容易,最佳富集纯化工艺为上样浓度5 g/L、上样pH值4、层析柱高径比7∶1、洗脱液为60%乙醇。使用该工艺对九节总黄酮进行纯化,精制后的总黄酮含量由纯化前17.6%提升至87.8%,约是纯化前的5倍。研究表明,试验获得了HP-20型大孔树脂吸附九节总黄酮的最佳纯化条件,为九节总黄酮作为饲料添加剂的开发利用提供参考。The study was to purify crude flavonoids from Psychotria asiatica by macroporous resin and determine the optimum purifying process for flavonoids.The study compared the adsorption and desorption performance of three macroporous resins including HP-20、AB-8 and D101,and select the best one to research the condition of sample and elution.Based on the single factor experiment,Box-Behnken response surface analysis was used to establish a mathematical model and optimize the purification conditions.The results showed that the optimum conditions for purification of crude extract of Psychotria asiatica with HP-20 macroporous adsorption resin were as follows:Loading concentration 5 g/L,loading pH value 4,ratio of height to diameter of macroporous resin column 7:1 and eluent volume fraction 60%.Under these conditions,the mass fraction of total flavonoids in Psychotria asiatica was increased from 17.6%before purification to 87.8%after purification,5 times higher after purification.The study indicate that HP-20 macroporous resin can be used to purify the total flavonoids in Psychotria asiatica,which can provide reference for the development and utilization of Psychotria asiatica as feed additives.
