摘  要:试验旨在使用超声波辅助优化博落回根原阿片碱、别隐品碱的提取工艺。采用单因素试验筛选提取工艺中的时间、液料比、提取次数的最佳条件,通过正交试验确定最佳提取条件;采用硅胶层析纯化原阿片碱、别隐品碱,使用液相色谱、红外光谱、紫外光谱表征所得产物。结果显示,液料比30 mL/g、提取1次、提取时间为30 min时,原阿片碱、别隐品碱提取率最高,分别为92.64%、90.93%。表征结果显示,提取物经柱层析纯化所得物质确定为别隐品碱和原阿片碱。研究表明,试验所用方法简单、快速,可为博落回资源的利用提供参考。The experiment was to optimize the extraction process of proopioid and allocryptinine from the root of Macleaya cordata with the aid of ultrasound.The optimal conditions of extraction time,liquid to solid ratio and extraction times were selected by single factor test,and the optimal extraction conditions were determined by orthogonal test.Proopioid and allocryptomine were purified by silica gel chromatography,and the products were characterized by liquid chromatography,infrared spectroscopy and ultraviolet spectroscopy.The results showed that the ratio of liquid to solid 30 mL/g,once extraction and the extraction time 30 min,the extraction rate of protoopium and allocryptomine was the highest,which were 92.64%and 90.93%,respectively.The results of characterization showed that the extracts purified by column chromatography were identified as allocryptomine and protoopioid.The experiment indicates that the method used in the experiment is simple and fast,which can provide reference for the utilization of the medicinal material resources.
