摘  要:目的探究姜黄素(CUR)对产后抑郁症(PPD)大鼠氧化应激损伤的影响及作用机制。方法将50只SD大鼠随机分为假手术组、PPD组、CUR低剂量(20 mg/kg)组、CUR高剂量(40 mg/kg)组、盐酸氟西汀(10 mg/kg)组,根据激素撤停法构建PPD大鼠模型,造模成功后分别给予不同药物处理。通过蔗糖偏好实验、强迫游泳测试、旷场实验评价大鼠抑郁程度;苏木精-伊红(H-E)染色观察海马组织病理学变化;试剂盒检测大鼠海马组织活性氧(ROS)、丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽(GSH)水平;Westernblot检测核因子E-2-相关因子2(Nrf2)/抗氧化反应元件(ARE)通路蛋白表达。结果与假手术组相比,PPD组大鼠海马组织结构受损,细胞排列松散,部分核固缩,蔗糖偏好度降低,游泳不动时间明显增加,旷场实验得分减少,ROS和MDA水平明显升高,SOD和GSH水平明显降低,Nrf2、HO-1蛋白水平显著降低,Keap1水平升高(均P<0.05);CUR低、高剂量组和盐酸氟西汀治疗后的PPD大鼠海马组织结构明显改善,细胞状态改善,蔗糖偏好度增加,游泳不动时间明显减少,旷场实验得分增加,氧化应激指标ROS和MDA水平明显降低,SOD和GSH水平升高,Nrf2、HO-1蛋白水平显著升高,Keap1水平下降(均P<0.05)。结论CUR可能通过激活Nrf2/ARE信号通路改善PPD大鼠氧化应激损伤,减轻PPD症状。Objective To explore the influence and mechanism of curcumin(CUR)on oxidative stress injury in rats with postpartum depression(PPD).Methods Fifty SD rats were randomly grouped into sham operation group,PPD group,low(20 mg/kg),high dose(40 mg/kg)CUR groups,and fluoxetine hydrochloride(10 mg/kg)group.The PPD rat model was constructed according to the hormone withdrawal method,and different drug treatments were given after the model was successfully established.The degree of depression in rats was evaluated by sucrose preference test,forced swimming test and open field test.Histopathological changes of hippocampus were observed by hematoxylin-eosin(H-E)staining.Kit was used to detect the levels of reactive oxygen species(ROS),malondialdehyde(MDA),superoxide dismutase(SOD)and glutathione(GSH)in rat hippocampus.Western blot was used to detect the expression of nuclear factor E-2-related factor 2(Nrf2)/antioxidant response element(ARE)pathway proteins.Results Compared with the sham operation group,the hippocampal tissue structure of the rats in the PPD group was damaged,the cells were loosely arranged,part of the nucleus was pyknosis,the sucrose preference was decreased,the swimming immobility time was greatly increased,and the open field test scores were decreased,ROS and MDA levels were greatly increased,SOD and GSH levels were greatly reduced,Nrf2 and HO-1 protein levels were greatly decreased,and Keap1 level was increased(all P<0.05).The hippocampal tissue structure of the PPD rats in the low and high dose CUR groups and after fluoxetine hydrochloride treatment was greatly improved,the cell state was improved,the sucrose preference was increased,the swimming immobility time was greatly reduced,and the open field test score was increased,ROS and MDA levels were greatly decreased,SOD and GSH levels were greatly increased,Nrf2and HO-1 protein levels were greatly increased,and Keap1 level was reduced(all P<0.05).Conclusion CUR may improve oxidative stress injury and alleviate PPD symptoms in PPD rats by activating the
