汉黄芩素调控lncRNA CDKN2B-AS1抑制宫颈癌细胞生长

摘　　要：目的探究汉黄芩素对宫颈癌细胞生长的作用机制。方法体外培养宫颈癌细胞株HeLa并分为对照组、低浓度组(汉黄芩素40μmol/L)、中浓度组(汉黄芩素80μmol/L)、高浓度组(汉黄芩素120μmol/L),采用MTT检测细胞增殖,采用流式细胞仪检测细胞凋亡,采用蛋白免疫印迹法检测Bax、Bcl-2、Caspase-3、Survivin蛋白表达量,采用RT-PCR法检测Bax、Bcl-2、Caspase-3、Survivin、lncRNA CDKN2B-AS1 mRNA表达变化。结果24 h、48 h、72 h时,低浓度组、中浓度组和高浓度组中宫颈癌细胞增殖指数、LncRNA CDKN2B-AS1 mRNA表达均低于对照组(P<0.05),有浓度依赖性。24 h时,高浓度组、中浓度组和低浓度组细胞凋亡率均高于对照组(P<0.05),有浓度依赖性;高浓度组Bax、Caspase-3蛋白和mRNA表达量高于对照组,Survivin、Bcl-2蛋白和mRNA表达量低于对照组。结论汉黄芩素可抑制宫颈癌细胞HeLa细胞增殖、促进其凋亡,还可抑制lncRNA CDKN2B-AS1表达。Objective To explore the action mechanism of wogonin on the growth of cervical cancer cells.Methods The cervical cancer cell lines HeLa were cultured in vitro and divided into control group,low-concentration(40μmol/L),medium-concentration(80μmol/L)and high-concentration wogonin groups(120μmol/L).The cells proliferation was detected by MTT.The cells apoptosis was detected by flow cytometry.The expressions of Bax,Bcl-2,Caspase-3 and Survivin proteins were detected by western blot.The changes in expressions of Bax,Bcl-2,Caspase-3,Survivin and lncRNA CDKN2B-AS1mRNA were detected by RT-PCR.Results At 24 h,48 h and 72 h,proliferation index of cervical cancer cells and expression of lncRNA CDKN2B-AS1 mRNA in low-concentration,medium-concentration and high-concentration groups were lower than those in control group(P<0.05),showing concentration dependence.At 24 h,apoptosis rate in low-concentration,mediumconcentration and high-concentration groups was higher than that in control group(P<0.05),showing concentration dependence.The expressions of Bax and Caspase-3 proteins and mRNA in high-concentration group were higher than those in control group,while expressions of Survivin and Bcl-2 proteins and mRNA were lower than those in control group.Conclusion Wogonin can inhibit the proliferation and promote the apoptosis of cervical cancer cells HeLa,which can also inhibit the expression of lncRNA CDKN2B-AS1.
