摘 要:目的:建立大秦艽汤(Daqinjiao Decoction,DQJD)HPLC指纹图谱及其9种成分的定量分析方法,为DQJD的质量控制标准的建立提供科学依据。方法:制备15批DQJD样品,以Venusil XBP C18(4.6 mm×250 mm,5μm)为色谱柱,流动相为0.1%磷酸水溶液(A)-乙腈(B),梯度洗脱,流速为1 mL·min-1,柱温30℃,进样体积5μL,检测波长237 nm,建立HPLC指纹图谱,采用“指纹图谱相似度评价系统软件(2012A版)”进行相似度分析,以马钱苷酸(loganic acid),龙胆苦苷(gentiopicroside)、芍药苷(paeoniflorin)、升麻素苷(cimicifugin)、阿魏酸(ferulic acid)、甘草苷(glycyrrhizin)、5-O-甲基维斯阿莱醇苷(5-O-methylvisalaitol glycoside)、黄芩苷(baicalin)、甘草酸铵(ammonium glycyrrhizinate)为指标性成分建立含量分析方法。结果:15批DQJD的指纹图谱相似度良好,确认了22个共有峰,9个指标成分分离度、精密度和重复性均良好,供试品溶液稳定性良好,9个指标成分具有良好的线性关系。结论:所建立的DQJD指纹图谱和含量测定方法简单可行、灵敏度高、重复性好,可用于DQJD的质量评价。OBJECTIVE To establish the HPLC fingerprint and quantitative analysis method for 9 components of Daqinjiao Decoction(DQJD),so as to provide scientific basis for the establishment of quality control standard of DQJD.METHODS Totally 15 batches of DQJD samples were prepared.A Venusil XBP C18(4.6 mm×250 mm,5μm)was used as a chromatographic column,the mobile phase was 0.1%phosphoric acid aqueous solution(A)-acetonitrile(B)with gradient elution,the flow rate was 1 mL·min-1,the column temperature was 30℃,and the injection volume was 5μL.The detection wavelength was 237 nm,and the HPLC fingerprint was established,and the Fingerprint Similarity Evaluation System Software(2012A version)was used for similarity analysis.Loganic acid,gentiopicroside,paeoniflorin,cimicifugin,ferulic acid,glycyrrhizin,5-O-methylvisalaitol glycoside,baicalin and ammonium glycyrrhizinate were used as the index components to establish a content analysis method.RESULTS The fingerprint similarity of 15 batches of DQJD was good,and totally 22 common peaks were confirmed.The resolution,precision and repeatability of 9 index components were promising,the stability of the test solution was high,and 9 index components had good linear relationship.CONCLUSION The established DQJD fingerprint and content determination method are simple and feasible with high sensitivity and good repeatability,which can be used for the quality evaluation of DQJD.
