摘  要:目的探讨缺氧诱导因子1α(hypoxia inducible factor 1α,HIF-1α)基因干扰对高糖(HG)诱导的肾小管上皮细胞(HK-2)凋亡、炎症和细胞外基质(ECM)沉积的影响。方法将HK-2细胞分为4组:对照组;HG组;HG+shRNA-NC组;HG+HIF-1α-shRNA组。按分组处理细胞后,实时定量聚合酶链反应检测HIF-1α和中性粒细胞明胶酶相关脂质运载蛋白(NGAL)的表达水平;细胞计数试剂盒8、流式细胞术和酶联免疫吸附试验分别检测细胞增殖、凋亡和炎性因子的含量;免疫印迹检测蛋白表达。结果与HG组相比较,HG+HIF-1α-shRNA组HIF-1α和NGAL mRNA和蛋白表达水平降低,细胞增殖倍数升高,细胞凋亡率降低,白细胞介素-6(IL-6)和肿瘤坏死因子-α水平降低,IL-10水平升高,组织金属蛋白酶抑制物l和I型胶原蛋白表达水平降低,基质金属蛋白酶9蛋白表达水平升高(P<0.01)。结论HIF-1α基因干扰缓解HG诱导的HK-2细胞凋亡、炎症和ECM沉积。Objective To investigate the effect of hypoxia-inducible factor 1α(HIF-1α)gene interference on high glucose(HG)-induced apoptosis,inflammation and extracellular matrix deposition in renal tubular epithelial cells(HK-2).Methods HK-2 cells were divided into 4 groups:control group,HG group,HG+shRNA-NC group,HG+HIF-1α-shRNA group.The expression levels of HIF-1αand neutrophil gelatinase-associated lipocalin(NGAL)were detected by quantitative real-time polymerase chain reaction.The cell proliferation,apoptosis and the levels of inflammatory factors were assayed by cell counting kit-8,flow cytometry and enzyme-linked immunosorbent assay,respectively.The protein expression levels were detected by Western blotting.Results Compared with the HG group,the mRNA and protein expression levels of HIF-1αand NGAL were decreased in the HG+HIF-1α-shRNA group,the cell proliferation rate was increased,the apoptosis rate was decreased,the levels of interleukin-6(IL-6)and tumor necrosis factor-αwere decreased,the level of IL-10 was increased,the expression levels of tissue inhibitors of metalloproteinase l and collagen type I protein were decreased,and the expression level of matrix metalloproteinase 9 protein was increased in the HG+HIF-1α-shRNA group(P<0.01).Conclusion HIF-1αgene interference alleviates HG-induced apoptosis,inflammation and extracellular matrix deposition in HK-2 cells.
