摘 要:目的探究软坚消瘿颗粒对实验性肝郁脾虚型桥本氏甲状腺炎(Hashimoto’s thyroiditis,HT)模型大鼠Th1/Th2型细胞因子表达的影响,为更进一步提高中医学治疗HT临床效果提供新思路和新方法。方法SD大鼠40只,随机取10只作为正常对照组,剩余大鼠采用高碘饮水与皮下注射甲状腺球蛋白联合方案建立桥本甲状腺炎大鼠模型,通过慢性束缚应激、过度疲劳、饮食失节复合方法建立肝郁脾虚大鼠模型。模型复制成功后,随机分为模型对照组(10只)、雷公藤多苷片组(10只)以及软坚消瘿颗粒组(10只)。正常对照组给予常规饲养,模型对照组予以蒸馏水2 mL,雷公藤多苷片组给予雷公藤多苷片9.45 mg/kg,软坚消瘿颗粒组给予软坚消瘿颗粒16.17 g/kg,各组干预时间为8周。干预结束后,比较各组大鼠一般情况、HE染色情况、甲状腺功能指标、细胞凋亡数、Th1/Th2型细胞因子表达情况。结果(1)正常组大鼠平均体质量为(322.69±38.92)g,模型对照组平均体质量为(206.77±21.02)g,雷公藤组平均体质量为(243.72±28.54)g,软坚消瘿组平均体质量为(280.53±34.54)g。与模型对照组、雷公藤组相比,软坚消瘿组平均体质量较高,差异计算有统计学意义(P<0.05)。(2)与模型对照组相比,雷公藤组与软坚消瘿组FT3、FT4、TRH水平较高,血清TSH、TGAb、TPOAb水平较低,差异计算有统计学意义(P<0.05);较雷公藤多苷片组相比,软坚消瘿颗粒组血清FT3、FT4、TRH水平较高,TSH、TGAb、TPOAb水平较低(P<0.05)。(3)与模型对照组相比,雷公藤组与软坚消瘿组甲状腺细胞凋亡数较低(P<0.05);与雷公藤多苷片组相比,软坚消瘿颗粒组甲状腺细胞凋亡数较低(P<0.05)。(4)与模型对照组相比,雷公藤组与软坚消瘿组Th1、Th17、Th1/Th2、Th17/Treg较低,Th2、Treg较高(P<0.05);与雷公藤多苷片组相比,软坚消瘿颗粒组Th1、Th17、Th1/Th2、Th17/Treg较低,Th2、Treg较高(P<0.05)。结论软坚消瘿�Objective To investigate the effect of Luanjian Xiaoying granules on the expression of Th1/Th2 cytokines in Hashimoto’s thyroiditis model rats with liver stagnation and spleen deficiency, and to provide new ideas and methods for further improving the clinical effect of Chinese medicine on HT. Methods Forty SD rats were randomly selected as normal control group(n=10). The remaining rats were treated with high-iodine drinking water combined with subcutaneous injection of thyroglobulin to establish Hashimoto’s thyroiditis rat model, and the liver stagnation and spleen deficiency rat model was established by chronic restraint stress, excessive fatigue and diet loss. After successful model replication, the rats were randomly divided into model control group(n=10), Tripterygium wilfordii polyglycoside tablet group(n=10) and Lujian Xiaoying granule group(n=10). The normal control group was given conventional feeding, the model control group was given 2mL distilled water, the Tripterygium wilforgium polyside tablet group was given 9.45 mg/kg tripterygium wilforgium polyside tablet, and the Luanjian Xiaoying granule group was given 16.17 g/kg Luanjian Xiaoying granule. The intervention time of each group was 8 weeks. After the intervention, the general conditions, HE staining, thyroid function indexes, the number of apoptosis, and the expression of Th1/Th2 cytokines were compared between the groups. Results(1) The average body weight of the normal group was(322.69±38.92) g, the average body weight of the model control group was(206.77±21.02) g, the average body weight of the tripterygium wilfordii group was(243.72±28.54) g, and the average body weight of the soft Jianxiao gall group was(280.53±34.54) g. Compared with the model control group and Tripterygium wilfordii group, the average body weight of the soft Jianxiao gall group was higher, and the difference was statistically significant(P<0.05).(2) Compared with model control group, the levels of FT3, FT4 and TRH in Tripterygium wilfordii and Lujianxiaogall grou
