龟羚帕安丸对帕金森病大鼠血清IL-6、IL-1β、TNF-α、Hcy、UA及APN水平的影响

摘　　要：目的观察龟羚帕安丸对帕金森病(Parkinson's disease,PD)大鼠血清白细胞介素-6(interleukin-6,IL-6)、白细胞介素-1β(interleukin-1β,IL-1β)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、同型半胱氨酸(homocysteine,Hcy)、尿酸(uric acid,UA)及脂联素(adiponectin,APN)水平的影响。方法PD大鼠模型采用6-羟基多巴胺(6-OHDA)注射法制作,造模成功的PD大鼠随机分为模型组,西药组,中药高、中、低剂量组,中西药合用组,每组15只,另设15只假手术组。模型组及假手术组给予等容积生理盐水灌胃,其余组给予相应药物灌胃,连续给药4周。大鼠腹主动脉取血,酶联免疫吸附测定法(ELISA)检测血清IL-6、IL-1β、TNF-α、Hcy、UA及APN水平。结果模型组大鼠血清IL-6、IL-1β、TNF-α及Hcy水平均明显升高,UA、APN水平均明显降低;假手术组、中药各剂量组、中西药合用组、西药组血清IL-6、IL-1β、TNF-α及Hcy水平均明显降低,UA、APN水平均明显升高。结论龟羚帕安丸具有明显神经保护作用,作用机制与降低IL-6、IL-1β、TNF-α及Hcy水平,增加UA、APN水平,发挥抗炎、抗氧化应激作用有关。Objective To observe the effects of Guiling Paan Pill(龟羚帕安丸)on the levels of serum interleukin-6(IL-6),interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α),homocysteine(Hcy),uric acid(UA)and adiponectin(APN)in Parkinson's disease(PD)rats.Methods The PD rat model was made by injection of 6-hydroxydopamine(6-OHDA).The successful PD rats were randomly divided into model group(n=15),western medicine group(n=15),Chinese medicine high dose group(n=15),Chinese medicine middle dose group(n=15),Chinese medicine low dose group(n=15)and combination group(n=15).The model group and sham operation group were given the same volume of normal saline,and the other groups were given the corresponding drugs for 4 weeks.The serum levels of IL-6,IL-1β,TNF-α,Hcy,UA and APN were detected by enzyme linked immunosorbent assay(ELISA).Results The levels of serum IL-6,IL-1β,TNF-αand Hcy in the model group were significantly increased,while the levels of UA and APN were significantly decreased,while the levels of serum IL-6,IL-1β,TNF-αand Hcy in the sham operation group,each dose group,combination group and western medicine group were significantly decreased,while the levels of UA and APN were significantly increased.Conclusion Guiling Paan Pill has obvious neuroprotective effect,and its mechanism is related to reducing the levels of IL-6,IL-1β,TNF-αand Hcy,increasing the levels of UA and APN,and exerting anti-inflammatory effect.
