摘  要:目的探讨清热解毒中药黄芩和栀子对肠道菌群的干预作用,为清热解毒中药基于肠道菌群治疗疾病提供理论依据。方法以高、中、低剂量黄芩和栀子灌胃大鼠,连续给药3周后收集大鼠十二指肠、空肠、回肠和结肠的肠段及内容物样本;采用实时荧光定量PCR法(qRT-PCR)检测各肠段肠球菌、大肠杆菌、拟杆菌、乳酸菌和梭菌的起始拷贝数,进而探讨这5种肠道主要优势菌的数量变化。结果在十二指肠:黄芩促进肠球菌的生长,抑制大肠杆菌、拟杆菌和乳酸菌的生长,对梭菌双向调节;栀子抑制大肠杆菌、乳酸菌和梭菌的生长,对肠球菌和拟杆菌双向调节。在空肠:黄芩促进肠球菌和乳酸菌生长,抑制拟杆菌和梭菌的生长,对大肠杆菌双向调节;栀子促进肠球菌和乳酸菌的生长,抑制大肠杆菌、拟杆菌和梭菌的生长。在回肠:黄芩抑制肠球菌、大肠杆菌、拟杆菌、乳酸菌和梭菌的生长;栀子:抑制肠球菌、大肠杆菌、拟杆菌、乳酸菌和梭菌的生长。在结肠:黄芩抑制肠球菌、大肠杆菌、拟杆菌和梭菌的生长,促进乳酸菌的生长;栀子抑制肠球菌、大肠杆菌、拟杆菌和梭菌的生长,对乳酸菌双向调节。结论黄芩和栀子对5种肠道优势菌的干预作用具有明显的共性和特征性,值得临床关注。Objective To investigate the intervention effect of Huangqin(Scutellariae Radix)and Zhizi(Gardeniae Fructus)on intestinal flora and provide theoretical basis for treating diseases based on intestinal flora.Method The rats were given high,medium or low dose of Huangqin(Scutellariae Radix)and Zhizi(Gardeniae Fructus)aqueous extracts by gavage.After three weeks of continuous administration,the intestinal segments and contents samples of the duodenum,jejunum,ileum and colon of the rats were collected.Then,qRT-PCR was used to detect the initial copy number of Enterococcus,Escherichia coli,Bacteroidetes,Lactobacillus and Clostridium in each intestinal segment,and then the number changes of the five dominant intestinal flora were discussed.Result In duodenum:Huangqin(Scutellariae Radix)promoted the growth of Enterococcus,inhibited the growth of Escherichia coli,Bacteroides and Lactobacillus,and regulated Clostridium bidirectionally.Zhizi(Gardeniae Fructus)inhibited the growth of Escherichia coli,Lactobacillus and Clostridium,and regulated the Enterococcus and Bacteroidetes bidirectionally.In jejunum:Huangqin(Scutellariae Radix)promoted the growth of Enterococcus and Lactobacillus,inhibited the growth of Bacteroides and Clostridium,and regulated the Escherichia coli bidirectionaliy.Zhizi(Gardeniae Fructus)promoted the growth of Enterococcus and Lactobacillus,and inhibited the growth of Escherichia coli,Bacteroides and Clostridium.In ileum:Huangqin(Scutellariae Radix)inhibited the growth of Enterococcus,Escherichia coli,Bacteroidetes,Lactobacillus and Clostridium.Zhizi(Gardeniae Fructus)inhibited the growth of Enterococcus,Escherichia coli,Bacteroidetes,Lactobacillus and Clostridium.In colon:Huangqin(Scutellariae Radix)inhibited the growth of Enterococcus,Escherichia coli,Bacteroidetes and Clostridium,and promoted the growth of Lactobacillus.Zhizi(Gardeniae Fructus)inhibited the growth of Enterococcus,Escherichia coli,Bacteroidetes and Clostridium and regulated Lactobacillus bidirectionally.Conclusion The intervention eff
