摘 要:为了解扬州市流浪犬相关肺炎克雷伯菌和大肠埃希菌的耐药情况及质粒介导的喹诺酮类耐药(plasmid-mediated quinolone resistant, PMQR)基因的流行性与多样性,从扬州市犬只留检所采集156份样品进行肺炎克雷伯菌和大肠埃希菌的分离、纯化与鉴定;采用琼脂稀释法测定分离株对15种抗菌药物的最小抑菌浓度,PCR检测喹诺酮类耐药菌株PMQR基因的携带情况。结果表明:共分离到82株肺炎克雷伯菌和119株大肠埃希菌,其中肺炎克雷伯菌对氨苄西林的耐药率最高(97.56%),其次是磷霉素(75.61%),有7株菌对6种以上的药物耐药;119株大肠埃希菌中有79株菌至少对1种药物耐药,对四环素、氨苄西林、萘啶酸、复方新诺明和环丙沙星的耐药率分别为48.74%、37.82%、26.05%、29.41%和10.08%, 12株菌耐6种以上药物。共分离到40株喹诺酮耐药菌株,PMQR基因qnrS、oqxAB、qnrD和aac(6′)-Ib-cr的检出率分别为22.50%、17.50%、7.50%和7.50%,且有1株肺炎克雷伯菌和1株大肠埃希菌同时携带aac(6′)-Ib-cr、qnrD和qnrS。综上,扬州市流浪犬是多重耐药菌潜在的储存库,且存在PMQR基因的流行,应引起重视。This study was conducted to investigate the antibiotic resistance of Klebsiella pneumoniae and Escherichia coli from stray dogs in Yangzhou and the prevalence and the diversity of plasmid-mediated quinolone resistance(plasmid-mediated quinolone resistant, PMQR) genes. In total, 156 samples were collected from Yangzhou Canine Inspection and Quarantine Institute in July 2021. K.pneumoniae and E.coli were isolated, purified and identified. The minimum inhibitory concentration of 15 antibacterial drugs was determined, and the PMQR genes were detected in all quinolone-resistant strains by PCR amplification. The results showed that 82 K.pneumoniae isolates and 119 E.coli isolates were detected, in which K.pneumoniae showed highest resistance to ampicillin(97.56%), followed by fosfomycin(75.61%), seven strains were resistant to more than six antibiotics. 79 of 119 E.coli isolates were resistant to at least one kind of antibiotic, and the resistance rates to tetracycline, ampicillin, nalidixic acid, cotrimoxazole and ciprofloxacin were 48.74%, 37.82%,26.05%,29.41% and 10.08%,respectively,and twelve isolates were resistant to at least six antibotics.Among the forty quinolone-resistant strains,the detection rates of PMQR genes qnrS,oqxAB,qnrD and aac(6’)-Ib-cr were 22.50%,17.50%,7.50% and 7.50%,respectively.One strain of K.pneumoniae and one strain of E.coli were detected to carry aac(6’)-Ib-cr,qnrDand qnrSgenes.In conclusion,stray dogs in Yangzhou may be a potential reservoir of multidrug-resistant bacteria,and the prevalence of PMQR gene should be concerned.
