摘  要:磷脂酰乙醇胺结合蛋白(phosphatidyl ethanolamine binding protein,PEBP)基因家族的氨基酸结构中存在保守的磷脂酰乙醇胺结合蛋白结构域,对调控植物开花起着至关重要的作用。本研究以染井吉野樱全基因组数据为基础,采用生物信息学方法鉴定出13个PEBP基因,依次命名为PYPEBP1~PYPEBP13,然后进一步分析了家族中各基因结构、理化性质,对PEBP蛋白的亚细胞定位、二级结构以及磷酸化位点进行预测,并构建系统发育树。结果表明,染井吉野樱的PEBP基因家族不同蛋白质序列的长度和特性都存在较大差异;13个染井吉野樱PEBP基因大部分成员含有4个外显子和3个内含子,motif 3、motif 4、motif 5和motif 6是染井吉野樱PEBP基因的特征基序;蛋白的亚细胞定位预测显示,这13个PEBP基因的蛋白分别位于细胞核与细胞质中;蛋白质二级结构分析显示,染井吉野樱PEBP基因的蛋白质二级结构十分相似,PYPEBP5和PYPEBP12蛋白结构中无规则卷曲占比最高,而β-转角比例最低;潜在的磷酸化位点预测分析表明,除PYPEBP3、PYPEBP7、PYPEBP8和PYPEBP10蛋白没有潜在的酪氨酸Tyr磷酸化位点外,其余PEBP蛋白均含有潜在的丝氨酸(Ser)、苏氨酸(Thr)和酪氨酸(Tyr)磷酸化位点,各蛋白中不同磷酸化位点的数量差异显著;系统发育分析表明,染井吉野樱PEBP基因与拟南芥PEBP基因亲缘关系最近,各个家族成员均可聚到一起。本研究结果为后续深入研究染井吉野樱PEBP基因家族的功能提供参考。There is a conserved domain of phosphatidylethanolamine binding protein in the amino acid structure of phosphatidylethanolamine binding protein gene family,which plays an important role in regulating plant flowering.Based on the whole genome data of Prunus yedoensis'Somei-yoshino',13 PEBP genes were identified by bioinformatics method,which were named PY PEBP1~PYPEBP13 in turn.Then,the structure and physicochemical properties of each gene in the family were further analyzed,and the subcellular location,secondary structure and phosphorylation site of PEBP protein were predicted,and the phylogenetic tree was constructed.The results showed that there were great differences in the length of different protein sequences and characteristics of PEBP gene family in P.yedoensis'Somei-yoshino'.Most of the 13 P.yedoensis'Somei-yoshino'PEBP genes contain 4 exons and 3 introns,and motif 3,motif 4,motif 5 and motif 6 were the characteristic motifs of PEBP genes of P.yedoensis'Somei-yoshino'.The subcellular localization prediction of proteins showed that the proteins of these 13 PEBP genes were located in the nucleus and cytoplasm respectively.The analysis of protein secondary structure showed that the protein secondary structure of PEBP gene of P.yedoensis'Somei-yoshino'was very similar,and the proportion of irregular curl was the highest in PYPEBP5 and PYPEBP12 protein structure,while the proportion of beta turn was the lowest.The analysis of potential phosphorylation sites showed that PEBP protein contained poten tial phosphorylation sites of serine(Ser),threonine(Thr)and tyrosine(Tyr)except PYPEBP3,PYPEBP7,PYPEBP8 and PYPEBP10,and the number of different phosphorylation sites in each protein was significantly different.Phylogenetic analysis showed that the PEBP gene of P.yedoensis'Somei-yoshino'was closely related to the PEBP gene of Arabidopsis thaliana,and all family members could get together.The results of this study provide reference for further study on the function of PEBP gene family of P.yedoensis Somei-yoshino'.
