摘　　要：TATA结合蛋白相关因子(TAF)是通用转录因子IID(TFIID)复合体中的一类亚基,它具有识别核心启动区域和激活转录起始的双重功能。拟南芥TAF的部分成员已被证实在光及激素信号转导、形态建成和生殖生长过程中发挥着重要的功能,但TAF9基因功能研究较少。本研究克隆了拟南芥TAF9的CDS及启动子,并对其基因功能和启动子活性进行了分析。结果发现:TAF9定位于细胞质和细胞核中;在长日照条件下该基因过表达显著延迟拟南芥的开花时间,且在TAF9过表达株系中,参与开花调控的关键基因CO、SPL3和FT的表达被抑制,而GA20ox1的表达被激活;从其启动子(-1500 bp)活性来看,TAF9主要在非种子组织中表达。本研究为TAF9的功能解析分析提供了重要参考。TATA-binding protein-associated factors(TAFs)are subunits of general transcription factor ID(TFIID)complex,and can recognize the core promoter of genes and activate transcription initiation.In Arabidopsis,some members of TAFs play important roles in phytohormone signal transduction,morphogenesis,and reproductive growth.While,the function of TAF9 was not uncovered.In this study,CDS and promoter of TAF9 were cloned and their function were investigated.TAF9 was localized in the nuclear and cytoplasm.The flowering time were later in the over-expression line of TA F9 than in Col-O under long-day culture condition.Moreover,the expression of key genes involved in flowering regulation,including CO,SPL3 and FT,was inhibited,while the expression of GA20ox1 was activated in TAF9 overexpressed lines.The promoter activity of TAF9(-1500 bp)was only detected in the non-seed tissues.Our results provided the basis for the function analysis of TA F9.
