摘 要:本研究以育成的‘云岭’系列水稻雌性核不育系为材料,利用水稻56K SNP芯片,构建水稻雌性核不育系SNP分子指纹数据库,同时检测分析其抗逆相关差异位点。结果表明,56K水稻SNP芯片共筛选出32727个标记位点,并利用12475个多态性SNP位点的物理位置构建了分子指纹图谱。籼粳组分分析发现10个雌性核不育系的粳型组分比例均大于籼型组分比例,‘云岭301FS’~‘云岭305FS’的籼型组分明显小于‘云岭306FS’~‘云岭310FS’,且籼粳位点差异明显。水稻雌性核不育系的抗逆相关基因主要有8种类型,共有41个基因,包含160个位点,表明供试的水稻雌性核不育系拥有良好的抵抗相应逆境的分子基础。通过比对参考基因组,发现一些基因的CDS区碱基发生不同程度的转换、颠换,其中有6个位点的改变导致了同义突变,13个位点的改变导致了错义突变,这可能导致多肽链原有功能的改变,最终影响水稻抵抗逆境的能力。本研究结果将为雌性核不育系的分子鉴定及其优良抗逆特性的研究提供理论基础。In this study,the Yunling'series of rice genic female sterile lines were used as materials and rice 56K SNP chips were used to construct the SNP molecular fingerprint database of rice genic female sterile lines,detect and analyze the different sites related to stress resistance.The results showed that the 56K rice SNP chip screened a total of 32727 marker loci,and constructed a molecular fingerprint using the physical location of 12475 polymorphic SNP loci.Analysis of indica and japonica components found that the ratio of japonica components in 10 genic female sterile lines was greater than that of indica components.The indica components of'Yunling301FS'~'Yunling305FS'were significantly smaller than that of'Yunling306FS'~'Yunling 310FS',and the difference between indica and japonica loci is obvious.There are 8 types of genes related to stress resistance of rice genic female sterile lines,with a total of 41 genes and 160 loci.It's indicates that the tested rice genic female sterile line has a good molecular basis to resist the corresponding adversity.By comparing the reference genome,it is found that the bases in the CDS region of some genes have undergone different degrees of conversion and transversion.Among them,the changes in 6 sites lead to synonymous mutations,and the changes in 13 sites lead to missense mutations.It may lead to changes in the original functions of the polypeptide chain,and ultimately affect the ability of rice to resist adversity.The results of this study will provide a theoretical basis for the molecular identification of rice genic female sterile lines and the study of stress resistances.
