摘  要:本研究发现以芥菜型褐籽油菜与芥菜型黄籽油菜进行杂交获得的F2后代中黄籽和褐籽后代的籽粒颜色并不是完全一致,黄籽有明亮的淡黄到暗黄的渐变,而褐籽有浅褐到深褐的渐变。因此,为了探究芥菜型油菜种皮颜色的机理,本研究以芥菜型褐籽油菜X398为母本,芥菜型黄籽油菜X402为父本进行杂交构建了重组自交系群体,再对重组自交系群体进行简化基因组测序,构建了芥菜型油菜高密度遗传连锁图谱,结合重组自交系群体的粒色性状表型和基因型进行QTL定位。结果获得4个控制芥菜型油菜黄籽性状的主效QTL,其中A09连锁群上获得1个主效QTL,其贡献率为14.2%,置信区间为47.6~49.6;B08连锁群上获得3个主效QTL,其贡献率分别为19.2%、12.9%和20.5%,置信区间分别为54.8~57.2、61.0~63.4和72.2~73.9。这为芥菜型油菜粒色性状基因的精细定位和分子标记辅助育种提供理论支撑。In this study,we found that the seed coat colors of both yellow and brown seeds of the F2 individuals from the cross between the brown-seed coat line and the yellow-seed coat line of B.juncea are not uniform.For example,there is a gradient from light yellow to dark yellow in the yellow seeds,while there is also a gradient from light brown to dark brown in the brown seeds.Therefore,in this study,in order to explore the causal mechanism that controls the seed coat color of B.juncea,the brown-seed coat line of B.juncea X398 was used as the female parent,and the yellow-seed coat line of B.juncea X402 was used as the male parent,we construct a recombinant inbred line(RIL)population.A high-density genetic linkage map was constructed by simplified genome re-se-quencing using RIL population.QTL mapping was conducted using the phenotype and genotype of the RIL population.As a result,four major QTLs for the yellow seed coat trait of B.juncea were obtained,and one of them was on the A09 chromosome,its contribution rate was 14.2%,interval was 47.6~49.6;and three of them were on the B08 chromosome,and their contribution rates were 19.2%,12.9%and 20.5%,interval was 54.8~57.2,61.0~63.4 and 72.2~73.9,respectively.This provides theoretical support for the fine mapping,cloning and molecular marker-assisted breeding of the grain color trait genes of B.juncea.
