摘  要:目的 探讨补肾活血方对慢性免疫性血小板减少症(chronic immune thrombocytopenia, CITP)的治疗作用及效应机制。方法 80只小鼠随机分为4组(对照组、ITP模型组、醋酸泼尼松组和补肾活血方组),采用被动免疫造模法建立CITP小鼠模型,第8天进行模型评估后,以补肾活血方、醋酸泼尼松进行药物干预14 d,采用酶联免疫吸附测定法(enzyme-linked immuno sorbent assay, ELISA)检测小鼠血管内皮细胞相关血管因子含量的影响。结果 与模型组比较,补肾活血方组、醋酸泼尼松组治疗14 d后,能显著提升血小板数目,显著降低前列环素(epoprostenol, PGI2)水平(P<0.05);而血浆血栓素(thromboxane, TXA2)、血小板激活因子(platelet activating factor, PAF)、血管内皮生长因子(vascular endothelial growth factor, VEGF)含量显著升高。补肾活血方组对TXA2、VEGF含量升高效果显著优于醋酸泼尼松组。结论 补肾活血方能够影响慢性免疫性血小板减少症模型小鼠血管内皮细胞相关血管因子TXA2、PGI2、PAF以及VEGR的水平,从而影响外周血小板数目,促进血管内皮细胞(vascular endothelial cell, VEC)修复,加强止血效应。而且,VEGF可作为评价ITP治疗及预后的指标。Objective To research and discussion the therapeutic effect and mechanism of Bushen Huoxue Decoction(补肾活血方)on chronic immune thrombocytopenia(CITP).Methods Eighty mice were randomly divided into 4 groups(control group,ITP model group,prednisone acetate group and Bushen Huoxue Decoction group).The CITP mouse model was established by passive immunomodeling.After the model was evaluated on the 8th day,the mice were treated with Bushen Huoxue Decoction and prednisone acetate for 14 days.The effects of the content of vascular endotheliacells(VEC)associated vascular factors were by ELISA.Results Compared with the model group,Bushen Huoxue Decoction group and prednisone group could significantly increase the number of platelets and decrease the level of cepoprostenol(PGI2)after 14 days of treatment(P<0.05),while the levels of thromboxane(TXA2),platelet activating factor(PAF),and vascular endothelia grouth factor(VEGF)were significantly increased.The effect of Bushen Huoxue Decoction on the increase of TXA2 and VECF was significantly better than prednisone.Conclusion Bushen Huoxue Decoction can affect the levels of TXA2,PGI2,PAF and VEGR related to VEC in mice with CITP model.thereby affecting the number of platelets,promoting the repair of VEC and strengthening the hemostatic effect.Moreover,VEGF can be used as an index to evaluate the treatment and prognosis of ITP.
