摘 要:目的 观察加味独活寄生合剂对兔膝骨关节炎软骨基质金属蛋白酶-3(MMP-3)、肿瘤坏死因子-α(TNF-α)表达的影响。方法 在成功制备兔KOA模型后,80只白兔随机分为正常对照组、模型对照组、阳性对照组和独活寄生组,各20只,独活寄生组给与加味独活寄生合剂灌胃,6.8 mL/(kg·d),阳性对照组给与盐酸氨基葡萄糖胶囊,0.07 g/(kg·d),正常对照组和模型对照组给与等剂量生理盐水灌胃,在实验干预前、干预第2、4、8周,免疫组织化学法检测膝关节炎软骨MMP-3、TNF-α表达,进行对比。结果 与模型对照组比较,独活寄生组关节软骨MMP-3、TNF-α在干预后第2周、第4周、第8周表达均减少,差异有统计学意义(P<0.05)。结论 加味独活寄生合剂可调控KOA关节软骨MMP-3、TNF-α表达,延缓软骨退变,这可能是其治疗KOA的作用机制。Objective To observe the effect of Jiawei Duhuo Jisheng Mixture(加味独活寄生合剂)on expressions of matrix metalloproteinase-3(MMP-3)and tumor necrosis factor-α(TNF-α)in rabbit with knee osteoarthritis(KOA).Methods After the successful preparation of the rabbit model of KOA,80 rabbits were randomly divided into normal control group,model control group,positive control group and Jiawei Duhuo Jisheng Mixture group,20 in each group.Rabbits in Jiawei Duhuo Jisheng Mixture groupwere given Jiawei Duhuo Jisheng Mixture[6.8 mL/(kg·d)].The positive control group was given glucosamine hydrochloride capsules[0.07 g/(kg:d)J.The normal control group and model control group were given isodose physiological saline by lavage.MMP-3 and TNF-αin articular cartilage were detected before the intervention,and on the 2nd,4th and 8th week after intervention.Results Compared with those of the model control group,the expressions of MMP-3 and TNF-αin the Jiawei Duhuo Jisheng Mixture group were reduced on the 2nd,4th and 8th week after intervention,and the differences were statistically significant(P<0.05).Conclusion Jiawei Duhuo Jisheng Mixture could reduce the expressions of MMP-3 and TNF-αin articular cartilage and delay the cartilage degeneration,which may be the mechanism of its treatment of KOA.
