摘　　要：目的 检测功能性便秘小鼠使用助阳通便膏治疗前后,结肠组织中水通道蛋白3(aquaporin 3,AQP3)表达的变化,探究助阳通便膏缓解功能性便秘与结肠组织中AQP3的关系。设计梯度浓度的助阳通便膏,为临床治疗浓度提供依据。方法 100只昆明小鼠,随机选取16只设为空白组,余下小鼠制备便秘模型(盐酸洛哌丁胺制备)。便秘小鼠随机分为模型组,助阳通便膏高、中、低浓度组,麻仁软胶囊对照组。各组应用相应药物连续灌胃14 d后观察疗效,应用免疫组化法观察各组小鼠结肠组织AQP3表达。结果 通过免疫组化染色法,与模型组相比,各组AQP3均有减少,差异具有统计学意义(P<0.05),其中助阳通便膏中浓度组小鼠粪便重量、粪便含水比、首次排黑便时间以及结肠组织中AQP3表达与空白组之间差异均无统计学意义(P>0.05),且与麻仁软胶囊对照组之间差异均不具有统计学意义(P>0.05)。结论 助阳通便膏可能是通过减少功能性便秘小鼠结肠组织中AQP3的异常表达,改善便秘。在设计的不同浓度组别中,助阳通便膏中浓度组效果最优。Objective To observe the changes in the expression of aquaporin 3(AQP3)in the colon tissue of mice with functional constipation before and after treatment with Zhuyang Tongbian Ointment(助阳通便膏)andexplore the relationship between the relief of functional constipation and AQP3 in the colon tissue.The gradient concentration of Zhuyang Tongbian Ointment was designed to provide basis for clinical treatment.Methods Of 100 Kunming mice,16 were randomly selected as the blank group,and the remaining mice were prepared as a constipation model(prepared by loperamide hydrochloride).Mice with constipation were randomly divided into model group,Zhuyang Tongbian Ointment high concentration group,Zhuyang Tongbian Ointment medium concentration group,Zhuyang Tongbian Ointment low concentration group,and Maren Soft Capsule(麻仁软胶囊)group.The therapeutic effects were observed after 14 days of continuous intragastric administration of the corresponding drugs in each group,and the expression of AQP3 in the colon tissue of the mice in each group was observed by immunohistochemical method.Results By immunohistochemical staining method,compared with that of the model group,the AQP3 of each group was reduced.The difference was statistically significant(P<0.05).There was no significant difference in fecal weight,fecal water content ratio,first black stool excretion time or AQP3 expression in colon tissue between theZhuyang Tongbian Ointment medium concentration group and the blank group(P>0.05),and there was no significant difference between the Zhuyang Tongbian Ointmentmedium concentration group and Maren Soft Capsule group(P>0.05).Conclusion Zhuyang Tongbian Ointment may improve constipation by reducing the abnormal expression of AQP3 in the colon tissue of mice with functional constipation.Among the designed different concentration groups,the Zhuyang Tongbian Ointmentmedium concentration grouphas the best effect.
