摘  要:目的探讨黄芪甲苷(AS-IV)对人间充质干细胞(MSCs)黏附、迁移、凋亡和分泌血管生成相关生长因子的影响,为深入研究AS-IV通过MSCs调控血管新生的作用机制奠定基础。方法将间充质干细胞随机分为实验组和对照组,实验组用300mg/L的AS-IV干预,对照组用等体积的PBS液处理。通过黏附能力测定实验、细胞划痕实验、流式细胞凋亡实验检测AS-IV对MSCs黏附、迁移和凋亡的影响;同时采用ELISA法检测两组MSCs分泌VEGF、Ang-1、Ang-2的表达。结果与对照组相比,实验组中细胞黏附数增多,细胞迁移宽度增加,细胞凋亡率下降,差异显著,均具有统计学意义(t=13.32、6.33,P<0.05);实验组MSCs分泌VEGF、Ang-1、Ang-2量均增多,差异显著,均具有统计学意义(t=6.10、9.41、8.18,P<0.05)。结论AS-IV能明显改善MSCs生物学功能,并促进血管生成相关生长因子分泌,具有较强血管新生潜能。Objective To study the effects of astragalosideIV(AS-IV)on the adhesion,migration,apoptosis and secretion of angiogenesis-related growth factors by human mesenchymal stem cells(MSCs),so as to lay the foundation for an in-depth study on the mechanism of the action of AS-IVregulating angiogenesis through MSCs.Methods MSCs were randomly divided into experimental and control groups.The experimental group was intervened with 300 mg/L of AS-IV,and the control group was treated with the equal volume of PBS solution.Adherent ability assay,cell scratch assayand flow apoptosis assay were used to detect the effects of AS-IV on the adhesion,migration and apoptosis of MSCs.The ELISA assay was also used to detect the expressions of secreted vascularendothlialgrowthfactor(VEGF),angiopoietin 1(Ang-1)and angiopoietin 2(Ang-2)in MSCs of both groups.Results Compared with those of the control group,the number of cell adhesion increased,the cell migration width increased and the apoptosis rate decreased in the experimental group.The differences were significant and all were statistically significant(P<0.05).Meanwhile,the levels of VEGF,Ang-1and Ang-2 secreted by MSCs in the experimental group were increased compared with those of the control group,and the differences were significant,all of which were statistically significant(P<0.05).Conclusion AS-IV can significantly improve the biological function of MSCs and promote the secretion of angiogenesis-related growth factors and it has a strong angiogenic potential.
