摘  要:目的评价不同配比黄芪、蜈蚣、守宫组成的“益气通络”药组对H22荷瘤小鼠肿瘤的抑制作用和免疫器官指数的影响,为“益气通络”药组的临床应用及其用于中药组方提供实验依据。方法建立腋下移植H22荷瘤小鼠模型,将实验动物随机分组,灌胃(ig)给药10 d后剥离瘤块,计算肿瘤抑制率;剥取小鼠肝脏、脾脏、胸腺,计算免疫器官指数,给药期间称体重,观察小鼠生存状态。动物实验分两批进行,第一批建模成功的小鼠42只随机分为7组,各组每天灌胃给药体积为0.2 mL:阴性对照组(H_(2)O,ig 0.2 mL/只)、阳性对照组(注射5-氟尿嘧啶(5-FU)30 mg·kg^(-1)/只,H_(2)O,ig 0.2 mL/只)、守宫蜈蚣1∶1实验组(守宫1,蜈蚣1 g·kg^(-1))、守宫蜈蚣3∶1实验组(守宫1.5,蜈蚣0.5 g·kg^(-1))、守宫蜈蚣6∶1实验组(守宫1.71,蜈蚣0.29 g·kg^(-1)),单用守宫组(守宫1 g·kg^(-1))和单用蜈蚣组(蜈蚣1 g·kg^(-1)),以明确守宫与蜈蚣较好的配比。第二批实验将小鼠36只(每天灌胃0.4 mL)随机分为阴性组、阳性组、第一批动物实验确定的守宫蜈蚣方较优配比组(守宫1.5,蜈蚣0.5 g·kg^(-1))以及“益气通络”药组低、中、高剂量组,其中“益气通络”药组低、中、高剂量组为守宫蜈蚣方较优配比再分别加入黄芪3.9、7.8、15.6 g·kg^(-1)。结果第一批动物实验结果表明单用守宫组、守宫与蜈蚣配比1∶1、3∶1、6∶1实验组的抑瘤率分别为29.9%,27.5%,59.6%,32.0%,守宫蜈蚣3∶1、6∶1组的脾指数较阳性对照组明显升高(P<0.01);说明守宫与蜈蚣质量比3∶1配伍最佳。第二批动物实验结果表明:与阴性对照组相比,守宫蜈蚣方较优配比组、“益气通络”药组低剂量组和高剂量组抑瘤率分别为65.02%、60.15%和68.89%,具有极其显著的统计学差异(P<0.01),但这3组组间无显著性差异,“益气通络”药组中剂量组抑瘤率为34.24%,具有显著性差异(P<0.05),高剂量组胸腺指数与Objective To evaluate the effect of the“benefiting Qi and dredging collateral”drug group composed of Huangqi(Astragali Radix),Wugong(centipede)and Shougong(gecko)in different proportions on tumor inhibition and immune organ index of H22 tumor bearing mice,so as to provide experimental basis for the clinical application of the“benefiting Qi and dredging collateral”drug group and its application in the formulation of traditional Chinese medicine prescriptions.Methods H22 tumor-bearing mouse model was established.After 10 days of administration,the tumors were separated and the tumor inhibition rate was calculated.The livers,spleensand thymus of the mice were stripped and the immune organ index was calculated.The mice living condition and their weights change were observed during the drug administration.Animal experiments were conducted in two batches,and the first batch of 42 successfully modeled mice were randomly divided into 7 groups,each group was administrated with the dose of 0.2 mL each day.The negative control group was injectedwith H2O0.2 mL.The positive control group was injected with 5-FU(30 mg·kg^(-1))and H_(2)O_(2)(0.2 mL).The Shougong(gecko)-Wugong(centipede)1:1 experimental group was given Shougong(gecko)(1 g·kg^(-1))and Wugong(centipede)(1 g·kg^(-1)).The Shougong(gecko)-Wugong(centipede)3:1 experimental group was given Shougong(gecko)(1.5 g·kg^(-1))and Wugong(centipede)(0.5 g·kg^(-1)).The Shougong(gecko)-Wugong(centipede)6:1 experimental group was given Shougong(gecko)(1.71 g·kg^(-1))and Wugong(centipede)(0.29 g·kg^(-1)).The Shougong(gecko)alone group was given Shougong(gecko)(1 g·kg^(-1))and the Wugong(centipede)alone group was given Wugong(centipede)(1 g·kg^(-1)).It was to clarify a better ratio of Shougong(gecko)to Wugong(centipede).The second batch of 36 successfully modeled mice(0.4 mL/d by gavage)were randomly divided into negative control group and positive control groupand the optimal ratio formula[Shougong(gecko)1.5 g·kg^(-1),Wugong(centipede)0.5 g·kg^(-1)],“benefiting
