摘  要:目的探讨腹部推拿对高脂饮食诱导肥胖大鼠摄食及ghrelin、GAS、PYY水平的影响及其作用机制。方法将SD雄性大鼠随机分为3组:正常组、模型对照组、腹部推拿组,每组10只。模型对照组、腹部推拿组采用高脂饲料喂养法复制单纯性肥胖动物模型。腹部推拿组给予腹部按法(按中脘穴)、腹部摩法(摩中脘穴)、腹部推法(自上腹往返推至下腹),上述手法共治疗15 min,以上治疗均为每天1次,连续干预28 d。模型对照组,只造模不干预。正常对照组,不造模不干预。但两组均每天束缚1次,每次10 min,连续28 d。干预结束后,检测三组大鼠摄食量、体质量、体长、Lee′s指数等评价其生理指标变化情况。采用放免技术检查大鼠下丘脑及胃组织中胃饥饿素((Ghrelin)、胃泌素(GAS)、神经肽Y NPY)的含量。应用免疫荧光检测方法观察下丘脑及胃组织Ghrelin表达情况。结果与正常组比较,模型对照组大鼠的摄食量、体质量、体长、Lee′s指数明显、脂肪系数、肝脏体比升高,胃、下丘脑组织中的ghrelin含量降低,GAS、NPY含量升高,差异有统计学意义(P<0.05)。与模型对照组比较,腹部推拿可以有效降低肥胖大鼠的体质量、(Lee′s指数等,降低脂肪在肝脏的沉积,差异有统计学意义P(<0.05)。同时,胃、下丘脑组织中的脑肠肽激素Ghrelin、GAS、NPY水平均有不同程度改善P<0.01,P<0.05)。Ghrelin的免疫荧光表达,腹部推拿组肥胖大鼠Ghrelin含量有所增加。结论腹部推拿一定程度上降低了肥胖模型大鼠的体质量、体脂量等,这可能与其影响单纯性肥胖大鼠下丘脑及胃肠道相关神经肽含量有关。但其深层次机制仍待进一步研究。Objective To investigate the effect of abdominal Tuina on the food intake and the levels of ghrelin,gastrin(GAS)and neuropeptide Y(NPY)in obese rats induced by high-fat diet and its mechanism.Methods The SD male rats were randomly divided into 3 groups:normal group,model control groupand abdominal Tuina group,with 10 rats in each group.The model control group and abdominal Tuina group were fed with high-fat feed to replicate the simple obesity animal model.The abdominal Tuina group was given abdominal pressing method[pressing Qihai(RN6)],abdominal rubbing method[rubbing Zhongwan(RN12)],abdominal pushing method(pushing back and forth from the upper abdomen to the lower abdomen).The above-mentioned manipulations were treated for a total of 15 minutes,and the above treatments were performed once a day,continuously for 28 days.After the intervention,the rats in the three groups were tested for food intake,body weight,body length and Lee′s index to evaluate the changes of their physiological indicators.The contents of ghrelin,GAS and NPY in hypothalamus and stomach tissues of rats were examined by radioimmunoassay.Ghrelin content in hypothalamus and gastric tissues was detected by immunofluorescence detection.ResultsCompared with those of the normal group,the food intake,body weight,body length,Lee′s index,fat coefficient and liver body ratio of the model control group rats were significantly increased,the ghrelin content in stomach and hypothalamus tissues was decreased,and the contents of GAS and NPY were increased with statistical significance(P<0.05).Compared with those of the model control group,abdominal Tuina can effectively reduce the body mass,Lee′s index and fat deposition in the liver of obese rats,with a statistically significant difference(P<0.05).At the same time,the levels of ghrelin,GAS and NPY in gastric and hypothalamic tissues were improved to varying degrees(P<0.01,P<0.05).Immunofluorescence showed the expression of ghrelin of obese rats in abdominal Tuina group increased.ConclusionAbdominal
