乌鲁木齐生牛乳中金黄色葡萄球菌的肠毒素检测及分离株同源性分析

摘　　要：探究乌鲁木齐生牛乳中金黄色葡萄球菌经典肠毒素携带情况及分离株同源性差异,为金黄色葡萄球菌的防控提供参考。对乌鲁木齐地区采集的131份生牛乳样品采用特异性选择培养基结合PCR方法分离鉴定金黄色葡萄球菌,并通过PCR方法检测金黄色葡萄球菌5种经典肠毒素(sea、seb、sec、sed、see)携带情况和根据测序结果对分离株进行同源性分析。结果显示,共分离鉴定到金黄色葡萄球菌13株,总分离率为9.92%(13/131);13株金黄色葡萄球菌经典肠毒素阳性菌株5株,阳性率为38.46%;13株金黄色葡萄球菌的同源性在93.7%~99.7%之间,遗传差异在0.3~6.6之间。研究表明,该地区生牛乳中存在一定金黄色葡萄球菌的污染,经典肠毒素阳性率较高,不同来源的分离株同源性高、差异性小。To explore the carrying situation of classical enterotoxin of Staphylococcus aureus in raw cow milk in Urumqi and the homologous differences among isolates,so as to provide reference for the prevention and control of Staphylococcus aureus,Staphylococcus aureus was isolated and identified by specific selective medium combined with PCR method from 131 raw milk samples collected in Urumqi.The classical enterotoxin carrying status of Staphylococcus aureus was detected by PCR,and the homology of the isolated strains was analyzed according to the sequencing results.The results showed that a total of 13 strains of Staphylococcus aureus were isolated and identified,and the total isolation rate was 9.92%.Among 13 strains of Staphylococcus aureus,5 strains were classical enterotoxin positive strains,and the positive rate was 38.46%.The homology of 13 strains of Staphylococcus aureus was between 93.7%and 99.7%,and the genetic difference was between 0.3 and 6.6,with high homology and little difference.This study showed that there was a certain contamination of Staphylococcus aureus in raw milk in this area,the positive rate of classical enterotoxin was high,and the genetic relationship of isolates from different sources was similar,and there was a certain degree of horizontal transmission of Staphylococcus aureus among dairy products.
