摘　　要：猪圆环病毒2型(Porcine circovirus 2,PCV2)感染引起的猪圆环病毒病(PCVD)严重危害世界养猪业,PCV2能够引起仔猪免疫抑制,其具体机制仍需要进一步探索。巨噬细胞是重要的免疫细胞,探索PCV2对巨噬细胞活化炎症小体的调控能够帮助进一步阐明PCV2致病机制。论文主要探讨PCV2感染对炎症小体活化的调控。分析了PCV2感染巨噬细胞对炎症小体通路的影响,用荧光定量PCR方法检测感染不同时期AIM2、caspase-1、IL-1β等的mRNA表达水平,利用蛋白免疫印迹法和ELISA法检测IL-1β蛋白水平的表达和上清的分泌,还检测了抑制AIM2表达对病毒感染介导炎性细胞因子产生的影响。通过上述研究发现,PCV2感染激活了AIM2炎症小体通路引发炎症细胞因子大量释放,推测其可能是PCV2引发过度炎症反应的机制。PCV2 infection can cause porcine circovirus disease(PCVD)which is a serious threat to porcine husbandry worldwide.PCV2 can cause immunosuppression in piglets,but the specific mechanism needs more investigation.Macrophages are important immune cells.Exploring the regulation of macrophage-activated inflammasome by PCV2 can help further understanding its pathogenesis.This study is to investigate the regulation of porcine circovirus 2(PCV2)infection mediated activation of inflammasome.In this study,how PCV2 infection affects the pathway of inflammasome was analyzed.The mRNA expression levels of AIM2,caspase-1 and IL-1βwere detected by qPCR at different infection time.The protein expression levels were detected by Western blot,and the supernatant IL-1βwas detected by ELISA.The effect of inhibiting AIM2 expression in PCV2 infection was studied to examine PCV2 mediated inflammatory cytokines.The results showed that PCV2 infection activated the AIM2 inflammasome and triggered the release of a large number of inflammatory cytokines,which was speculated to be invovled in the excessive inflammatory response to PCV2 infection.
