摘 要:目的:探讨经皮穴位电刺激对阿尔茨海默病(AD)大鼠学习记忆能力、氧化应激和神经元凋亡的影响。方法:SPF级SD雄性大鼠48只,随机分为空白组(Control)、模型组(AD)、电针组(AD+EA)和经皮穴位电刺激组(AD+TEAS),每组12只。空白组颈背部皮下注射生理盐水,其余各组颈背部皮下注射D-半乳糖联合Aβ_(1-42)建立AD大鼠模型。空白组与模型组正常喂养,定时抓取以及固定于鼠套中20 min/d,但不给予干预;电针组给予电针干预;经皮穴位电刺激组给予经皮穴位电刺激干预。各组均连续干预4周。Morris水迷宫检测大鼠空间学习记忆能力;尼氏染色检测海马神经元损伤状况;酶联免疫吸附法(ELISA)检测大鼠皮层超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(Malondialdehyde,MDA)的表达;实时荧光定量PCR和蛋白质印迹法(Western blot)分析检测大鼠海马、皮层组织中氧化应激与凋亡相关表达。结果:与电针组相比,经皮穴位电刺激组治疗效果更好,AD大鼠找到平台的时间缩短,穿越平台次数增加,停留在目标象限的时间延长;且经皮穴位电刺激治疗后显著增加了AD模型大鼠海马区尼氏小体的数量且病理结构明显改善,增加抗氧化应激产物,下调凋亡因子和氧化应激的表达,上调了抗凋亡因子和抗氧化还原保护因子的表达。结论:经皮穴位电刺激治疗可能通过抗氧化应激,抑制神经元凋亡,从而改善大鼠的学习记忆能力。Objective:To investigate the effect of transcutaneous electrical acupoint stimulation(TEAS)on learning and memory ability and oxidative stress in rats with Alzheimer′s disease(AD).Methods:48 SD rats were randomly divided into the blank group,the model group,the EA group and the TEAS group,with 12 rats in each group.AD rat model was induced by subcutaneous injection of D-galactose combined with Aβ_(1-42).The EA group was given electro-acupuncture intervention,the TEAS group was given TEAS intervention,but the blank group and the model group were not given any intervention but 20 min/d fixing.After 4 weeks of corresponding intervention,Morris water maze was used to detect the spatial learning and memory ability in rats.Nissl staining was used to detect the damage of hippocampal neurons.ELISA method was used to detect the expressions of SOD and MDA in rat cortex.The expressions of oxidative stress and apoptosis in hippocampus and cortex were detected by real-time fluorescent quantitative PCR and Western blot.Results:Compared with those in the EA group,the effect was more significant,the time of finding platform was shorter,the number of platform crossing was increased,the time of staying in the target quadrant was prolonged in the TEAS group.TEAS treatment significantly increased the Nissl bodies in hippocampus of AD rats,and obviously improved pathological structure,increased the products of anti-oxidative stress,downregulated the expressions of apoptosis and oxidative stress,up-regulated the expression of anti-apoptotic factors and anti-redox protective factors.Conclusion:TEAS may improve learning and memory ability of AD rats by reducing oxidative stress and inhibiting neuronal apoptosis.
