摘　　要：目的建立同时测定除湿止痒软膏中9种成分含量的高效液相色谱(HPLC)法。方法色谱柱为Sunfire ODS C_(18)柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.1%磷酸溶液(梯度洗脱),流速为1.0 mL/min,检测波长为290 nm(0~15 min,虎杖苷和黄柏碱),220 nm(15~50 min,氧化苦参碱、苦参碱、小檗碱、黄柏酮、梣酮),322 nm(50~60 min,蛇床子素),254 nm(60~70 min,大黄素),柱温为30℃,进样量为10μL。结果蛇床子素、小檗碱、黄柏碱、黄柏酮、梣酮、苦参碱、氧化苦参碱、虎杖苷、大黄素的质量浓度分别在4.88~244.0μg/mL(r=0.9998)、2.86~143.0μg/mL(r=0.9996)、1.16~58.0μg/mL(r=0.9987)、1.42~71.0μg/mL(r=0.9991)、0.58~29.0μg/mL(r=0.9995)、0.86~43.0μg/mL(r=0.9994)、0.96~48.0μg/mL(r=0.9989)、1.84~92.0μg/mL(r=0.9997)、3.06~153.0μg/mL(r=0.9993)范围内与峰面积线性关系良好;精密度、稳定性和重复性试验结果的RSD均小于2.0%;平均加样回收率分别为99.37%,99.77%,99.56%,99.48%,98.98%,98.82%,100.93%,98.94%,100.64%,RSD分别为0.72%,0.98%,1.21%,1.16%,1.59%,1.13%,1.34%,0.82%,1.20%(n=9)。结论该方法操作简便,结果准确可靠,可用于同时测定除湿止痒软膏中9种成分的含量。Objective To establish a high-performance liquid chromatography(HPLC)method for the simultaneous determination of nine components in Chushi Zhiyang Ointment.Methods The chromatographic column was Sunfire ODS C_(18) column(250 mm×4.6 mm,5μm),the mobile phase was acetonitrile-0.1%phosphoric acid solution(gradient elution),the flow rate was 1.0 mL/min,the detection wavelength was set 290 nm(0-15 min,polydatin and phellodendrine),220 nm(15-50 min,oxymatrine,matrine,berberine,obacunone and fraxinellone),322 nm(50-60 min,osthole),and 254 nm(60-70 min,emodin),the column temperature was 30℃,and the injection volume was 10μL.Results The linear ranges of osthole,berberine,phellodendrine,obacunone,fraxinellone,oxymatrine,matrine,polydatin and emodin were 4.88-244.0μg/mL(r=0.9998),2.86-143.0μg/mL(r=0.9996),1.16-58.0μg/mL(r=0.9987),1.42-71.0μg/mL(r=0.9991),0.58-29.0μg/mL(r=0.9995),0.86-43.0μg/mL(r=0.9994),0.96-48.0μg/mL(r=0.9989),1.84-92.0μg/mL(r=0.9997),and 3.06-153.0μg/mL(r=0.9993),respectively.The RSDs of precision,stability,and repeatability tests were lower than 2.0%(n=9).The average recoveries of the above nine components were 99.37%,99.77%,99.56%,99.48%,98.98%,98.82%,100.93%,98.94%and 100.64%with RSDs of 0.72%,0.98%,1.21%,1.16%,1.59%,1.13%,1.34%,0.82%and 1.20%(n=9),respectively.Conclusion The method is simple,accurate,and reliable,which can be used for the simultaneous determination of nine components in Chushi Zhiyang Ointment.
