摘  要:目的探讨滋补脾阴方药对脾阴虚大鼠骨骼肌线粒体呼吸链复合物及ATP活性的影响。方法将Wistar大鼠随机分为正常组、脾阴虚组和滋补脾阴方药组,采用经典复合因素造模法(饮食失节、劳倦过度结合耗伤阴液法)建立脾阴虚大鼠模型,使用细胞能量代谢分析仪检测骨骼肌线粒体呼吸链复合物Ⅰ、Ⅱ、Ⅲ、Ⅳ及ATP合酶活性变化及滋补脾阴方药的干预作用。结果与正常组相比较,脾阴虚组大鼠骨骼肌线粒体呼吸链复合物Ⅰ、Ⅱ、Ⅲ、Ⅳ活性均上升(P<0.01),ATP合酶活性增加(P<0.001);与脾阴虚组相比,滋补脾阴方药组大鼠骨骼肌线粒体呼吸链复合物Ⅰ、Ⅱ、Ⅲ、Ⅳ活性均下降(P<0.01),ATP合酶活性减低(P<0.001)。结论滋补脾阴方药能够改善脾阴虚大鼠骨骼肌线粒体能量代谢异常,其机制可能与调节线粒体呼吸链复合物Ⅰ、Ⅱ、Ⅲ、Ⅳ及ATP合酶的活性有关。Objective To investigate the effect of Zibu Piyin Decoction(滋补脾阴方,ZBPYD)on mitochondrial respiratory chain complex and ATP activity in skeletal muscle of spleen Yin deficiency rats.Methods Wistar rats were randomly divided into normal group,spleen Yin deficiency group and ZBPYD group.The rat model of spleen Yin deficiency was established by classical compound factor modeling(diet disjunction,excessive tiredness combined with consumption of Yin-fluid method).The activities of mitochondrial respiratory chain complexesⅠ,Ⅱ,Ⅲ,Ⅳand ATP synthase in skeletal muscle and the intervention effect of ZBPYD were detected by cell energy metabolism analyzer.Results Compared with the normal group,the activities of respiratory chain complexesⅠ,Ⅱ,ⅢandⅣin skeletal muscle mitochondria of rats in the spleen Yin deficiency group were increased(P<0.01),and the ATP synthase activity was increased(P<0.001).Compared with the spleen Yin deficiency group,the activities of respiratory chain complexesⅠ,Ⅱ,ⅢandⅣin skeletal muscle mitochondria of rats in ZBPYD group were decreased(P<0.01),and the activity of ATP synthase was significantly reduced(P<0.001).Conclusion ZBPYD can improve the abnormal mitochondria energy metabolism of skeletal muscle in spleen Yin deficiency rats,and the mechanism may be related to the regulation of mitochondrial respiratory chain complexesⅠ,Ⅱ,Ⅲ,Ⅳand ATP synthase.
