摘  要:目的基于数据挖掘和网络药理学研究专利文献中改善失眠类中药复方用药规律及其作用机制。方法检索国家知识产权局、知网等数据库中改善失眠类中药专利复方,运用编程语言Python3对其进行用药频次、应用剂型、适宜证型统计及关联规则和聚类分析,获取用药规律及核心配伍结构。再利用网络药理学方法对核心配伍结构改善失眠作用机制进行进一步的探索。结果(1)数据挖掘纳入专利复方1047首,涉及中药852味;频次>100的中药有33味,其中以补虚、安神用药为主;通过用药频次、关联规则及聚类分析综合获得4味药物的核心配伍结构;用药剂型以传统剂型汤剂为主;适宜证型以肝火扰心证、心脾两虚证出现频次最多。(2)核心配伍结构有效活性成分79个,对应靶基因829个,交集靶基因140个;其核心作用靶蛋白有APP、TNF、AKT1、IL6等26个;KEGG通路富集分析主要涉及神经系统调节、炎症反应及能量代谢多条信号通路。结论中药专利复方在改善失眠方面符合补虚泻实、调整阴阳的治疗方法。酸枣仁-远志-茯苓-首乌藤可能通过神经系统调节、炎症反应及能量代谢多条通路发挥改善睡眠作用。研究结果可为基于中药专利信息促进改善失眠类中药产品的进一步开发提供一定的理论参考。Objective Based on data mining and network pharmacology to explore the medication rules and action mechanism of traditional Chinese medicine compound for improving insomnia in patent literature.Methods Searching Chinese medicine patent compound for improving insomnia in databases such as the State Intellectual Property Office and CNKI.Python 3 was used to analyze the frequency of medication,dosage forms,statistics of appropriate syndrome types,association rules and cluster analysis,and to obtain the medication rules and core compatibility structure.Then,the network pharmacology method was used to further explore the mechanism of core compatibility structure for improving insomnia.Results(1)1047 patent compounds were included,involving 852 Chinese medicines in data mining.There were 33 kinds of traditional Chinese medicines with frequency greater than 100 times,among which tonifying deficiency and calming nerves were the main drugs.The core compatibility structure of the four drugs was obtained by drug frequency,association rules and cluster analysis,the main dosage form was traditional decoction,and the frequent occurrence of syndrome types were liver-fire disturbing heart and deficiency of both heart and spleen.(2)There were 79 active components in core compatibility structure,corresponding to 829 target genes,140 intersecting target genes and 26 core target proteins such as APP,TNF,AKT1 and IL6.KEGG pathway enrichment analysis mainly involveed multiple signal pathways of nervous system regulation,inflammatory response and energy metabolism.Conclusions The patent compound of traditional Chinese medicine followes the treatment methods that tonifying deficiency and purging excess and adjusting Yin and Yang in improving insomnia.Suanzaoren(Ziziphi Spinosae Semen)-Yuanzhi(Polygalae Radix)-Fuling(Poria)-Shouwuteng(Polygoni Multiflori Caulis)may play a role in improving sleep through multiple pathways of nervous system regulation,inflammatory response and energy metabolism.The research results can provide a theoretica
