摘 要:具有高生物相容性和生物活性的荧光材料在创伤修复、药物递送、传感器、生物测定与成像等许多领域因便于示踪或实时监测而备受青睐。对丝素缺失突变型家蚕添食荧光染料罗丹明B,系统地研究添食罗丹明B对家蚕生理指标和茧质的影响,同时研究罗丹明B在家蚕体内的转移,以及添食后丝腺中丝胶液和蚕茧中罗丹明B的含量,制备彩色荧光丝胶纤维支架并分析其细胞相容性。结果表明,添食罗丹明B后家蚕幼虫体色、蚕茧或纤维支架均呈粉红色,随着罗丹明B添加量的增加,罗丹明B在家蚕体内存留量也随之提高,6 mg/mL罗丹明B处理组家蚕丝腺中丝胶液和蚕茧中的罗丹明B含量分别为0.011 mg/mL和0.166 mg/g;添食罗丹明B对家蚕的生命力和茧质均有显著影响,随着罗丹明B添加量的增加,家蚕的生命力和茧质均呈现下降趋势;添食获得的含有罗丹明B的蚕茧或纤维支架具有强荧光特性;添食罗丹明B可获得平整、均一的彩色荧光丝胶纤维支架,且该荧光支架无细胞毒性。综上,对丝素缺失突变型家蚕添食荧光色素是获得新型荧光丝胶材料的可行性途径。Fluorescent materials with high biocompatibility and bioactivity are favored in many fields such as wound repair,drug delivery,sensors,biometrics,and imaging for their ease of tracking or real-time monitoring.This research systematically studied the effects of feeding rhodamine B on physiological indexes and cocoon quality of fibroin-deficient silkworm.At the same time,the transfer of rhodamine B in silkworm,the content of rhodamine B in sericin fluid of silk gland and in cocoon after feeding were studied as well.The colored fluorescent sericin fibrous scaffolds were prepared and studied for their cytocompatibility.The results showed that after feeding rhodamine B,the body color of larvae,and the color of silkworm cocoons and fibrous scaffolds were all pink.With the increase of rhodamine B addition,the amount of rhodamine B retained in the silkworm body also increased.In the 6 mg/mL rhodamine B treatment group,the content of rhodamine B in sericin fluid of silk gland and in silkworm cocoon were 0.011 mg/mL and 0.166 mg/g,respectively Feeding rhodamine B had a significant effect on the viabilty and cocoon quality of silkworm.The viability and cocoon quality of silkworm both decreased with the increase of rhodamine B addition.Silkworm cocoon or fibrous scaffold containing rhodamine B obtained by feeding had strong fluorescence properties.Feeding rhodamine B can obtain a flat,uniform and colored fluorescent sericin fibrous scaffold without cytotoxicity.In conclusion,the addition of fluorescent pigments to fibroin-deficient silkworm is a feasible way to obtain new fluorescent sericin materials.
