摘 要:目的观察不同浓度双酚A(BPA)染毒模型与氢化可的松诱导肾阳虚模型小鼠睾丸细胞凋亡及雄激素受体(AR)和雄激素受体磷酸化(p-AR)定位与表达,探讨BPA与肾阳虚导致雄性生殖生精障碍的相关性。方法选取25只SPF级6周龄雄性昆明小鼠作为研究对象。根据不同制备方式分为1mg/kg BPA模型组、10mg/kg BPA模型组、100mg/kg BPA模型组、肾阳虚组和正常对照组。采用TUNEL染色观察睾丸组织细胞凋亡,免疫组化染色、免疫荧光染色及Western blotting观察AR、p-AR定位与表达。结果随着BPA浓度升高,BPA染毒模型小鼠睾丸生精细胞凋亡明显增加,AR睾丸表达显著下降,p-AR表达显著上升(P<0.05),且100mg/kg BPA组与肾阳虚组间比较,差异无统计学意义(P>0.05)。结论100mg/(kg·d)BPA和肾阳虚可以通过抑制AR表达、激活AR磷酸化来诱导小鼠睾丸生精细胞凋亡,这为进一步探讨补肾阳中药治疗BPA染毒导致的男性生精功能障碍提供了可靠的实验依据。Objective To observe the apoptosis of testicular cells and the localization and expression of androgen receptor(AR)and phosphorylated androgen receptor(p-AR)in mice exposed to different concentrations of bisphenol A(BPA)and hydrocortisone induced kidney-Yang deficiency models,and to explore the correlation between BPA and male reproductive and spermatogenic disorders caused by kidney-Yang deficiency.Methods 25 SPF-grade,6 week-old male Kunming mice were selected as the study subjects.According to different preparation methods,they were divided into 1 mg/kg BPA model group,10 mg/kg BPA model group,100 mg/kg BPA model group,kidney-Yang deficiency group,and normal control group.TUNEL staining was used to observe apoptosis in testicular tissue,and immunohistochemical staining,immunofluorescence staining,and Western blotting were used to observe the localization and expression of AR and p-AR.Results With the increase of BPA concentration,the apoptosis of spermatogenic cells in the testis of mice exposed to BPA significantly increased,the expression of AR in the testis significantly decreased,and the expression of p-AR significantly increased(P<0.05).There was no significant difference between the 100 mg/kg BPA group and the kidney-Yang deficiency group(P>0.05).Conclusions 100 mg/(kg·d)BPA and kidney-Yang deficiency can induce apoptosis of mouse testicular spermatogenic cells by inhibiting AR expression and activating AR phosphorylation,which provides a reliable experimental basis for further exploring the treatment of male spermatogenic dysfunction caused by BPA exposure with traditional Chinese medicine for tonifying kidney-Yang.
