黄芩苷对宫颈癌细胞系C33A增殖和凋亡的影响

摘  要:目的探讨黄芩苷影响宫颈癌细胞C33A增殖、凋亡的机制。方法采用CCK-8法检测经不同浓度黄芩苷处理的宫颈癌细胞系C33A的增殖情况;Annexin V-PI双染法检测经黄芩苷处理的宫颈癌细胞系C33A的凋亡情况;实时荧光定量聚合酶链反应(qRT-PCR)法检测经不同浓度黄芩苷处理的宫颈癌细胞系C33A Bcl-2、Bax、survivin和c-Myc基因相对表达量。结果黄岑苷能够抑制宫颈癌细胞系C33A的增殖,当黄芩苷浓度达到40μg/mL时,抑制作用达最大。24h、48h、72h半抑制浓度(IC_(50))分别为20.22μg/mL、19.81μg/mL、20.9μg/mL;黄芩苷在一定浓度范围内促进细胞系C33A凋亡的作用呈现浓度依赖性(P<0.05)。实验组Bcl-2、c-Myc、survivin基因表达量低于对照组,且呈剂量依赖性递减;而Bax基因表达量高于对照组(P<0.05)。结论黄芩苷可通过上调Bax的表达、下调Bcl-2、c-Myc和survivin的表达抑制宫颈癌细胞系C33A的增殖,并促进其凋亡。Objective To investigate the effect of baicalin on proliferation and apoptosis of human cervical cancer cells C33A.Methods CCK-8 method was used to detect the effect of baicalin on the proliferation of cervical cancer C33A cells.Apoptosis of C33A cells treated with baicalin was detected by Annexin V-PI double staining.The relative gene expression levels of Bcl-2,Bax,survivin and c-Myc in C33A cells treated with different concentrations of baicalin were detected by qRT-PCR.Results Flavin could inhibit the proliferation of cervical cancer cell line C33A,and the inhibitory effect reached the maximum when the concentration of baicalin reached 40μg/mL.The 50%concentration(IC_(50))at 24h,48h and 72h were 20.22μg/mL,19.81μg/mL and 20.9μg/mL,respectively.Baicalin promoted cell line C33A apoptosis in a concentration dependent manner(P<0.05).The expressions of Bcl-2,c-Myc and survivin genes in the experimental group were lower than those in the control group,and decreased in a dose-dependent manner.The expression level of Bax gene was higher than that of control group(P<0.05).Conclusions Baicalin can inhibit C33A proliferation and promote apoptosis of cervical cancer cells by up-regulating the expression of Bax and down-regulating the expression of Bcl-2,c-Myc and survivin.
