摘 要:目的探究环状RNA PRMT5(circPRMT5)对乳腺癌MCF-7细胞增殖、凋亡、侵袭的影响。方法采用qRT-PCR检测乳腺癌组织和细胞中circPRMT5的表达;shRNA-NC和circPRMT5-shRNAs转染至MCF-7细胞后,应用qRT-PCR检测circPRMT5的相对表达水平;克隆形成实验、流式细胞术、Transwell和划痕实验分别检测细胞增殖、凋亡、侵袭和迁移能力;Western blot法检测Ki-67、Survivin、cleaved caspase-3、cleaved caspase-9、基质金属蛋白酶-2(MMP-2)和MMP-9的蛋白表达水平。体内异种移植瘤实验验证circPRMT5在肿瘤生长中的作用。结果circPRMT5在乳腺癌组织和细胞中高表达(P<0.05)。与对照组相比,干扰circPRMT5表达能抑制MCF-7细胞增殖、侵袭和迁移,促进细胞凋亡(P<0.05)。在体内实验中,与shRNA-NC组比较,circPRMT5-shRNA1组肿瘤体积和肿瘤质量显著减小,Ki-67阳性细胞数显著减少,而caspase-3阳性细胞数显著增多(P<0.05)。结论干扰circPRMT5可抑制乳腺癌细胞增殖、迁移和侵袭,并促进细胞凋亡。Objective To investigate the effect of circular RNA PRMT5(circPRMT5)on proliferation,apoptosis and invasion of breast cancer MCF-7 cells.Methods circPRMT5 expression in breast cancer tissues and cells was detected by qRT-PCR.circPRMT5 expression was detected by qRT-PCR after transfection of shRNA-NC and circPRMT5-shRNAs into MCF-7 cells.Cell proliferation,apoptosis,invasion and migration were detected by clonal formation assay,flow cytometry,Transwell assay and scratch assay,respectively.The protein expression levels of Ki-67,Survivin,cleaved caspase-3,cleaved caspase-9,matrix metalloproteinase-2(MMP-2),and MMP-9 were detected by Western blot.The effect of circPRMT5 on tumor growth was verified by xenotransplantation in vivo.Results circPRMT5 was highly expressed in breast cancer tissues and cells(P<0.05).Compared with the control group,interfering the expression of circPRMT5 could inhibit cell proliferation,invasion and migration,and promote cell apoptosis(P<0.05).In vivo,compared with the shRNA-NC group,the tumor volume and mass of circPRMT5-shRNA1 group were significantly reduced,the number of Ki-67 positive cells was significantly reduced,and the number of caspase-3 positive cells was significantly increased(P<0.05).Conclusion Interfering circPRMT5 can inhibit the proliferation,migration and invasion of breast cancer cells and promote apoptosis.
