基于JAK2/STAT3信号通路探讨参茸地黄汤调控自噬改善糖尿病的作用机制

摘　　要：目的探讨参茸地黄汤干预JAK2/STAT3信号通路调控自噬改善糖尿病的作用机制。方法8周龄SD雄性大鼠,高脂饲料喂养4周后联合链脲佐菌素40 mg·kg^(-1)单次腹腔注射诱导2型糖尿病大鼠动物模型,中药组予参茸地黄汤低、高剂量,西药组予盐酸二甲双胍。观察大鼠的一般状态、体质量、饮水量,各组SD大鼠连续灌胃4周后麻醉处死取材,检测各组大鼠空腹血糖(FBG)、糖化血清蛋白(GSP)、甘油三酯(TG)、胆固醇(CHO)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、游离脂肪(FFA),空腹血清胰岛素(INS)。HE染色观察SD大鼠肝脏组织病理形态改变,Western blot法检测JAK2、STAT3、Beclin-1、LC3、GLUT4蛋白的表达。结果参茸地黄汤可改善2型糖尿病SD大鼠一般状态,改善体质量下降趋势,改善糖、脂代谢,并且可以提高JAK2、STAT3蛋白表达水平,降低Beclin-1、LC3Ⅱ/Ⅰ蛋白表达水平,提高GLUT4蛋白表达。结论参茸地黄汤治疗2型糖尿病可能是通过干预JAK2/STAT3信号通路,介导细胞自噬,提高GLUT4表达水平,改善胰岛素抵抗实现的。Objective Discuss the mechanism by which Shenrong Dihuang Decoction interferes with JAK2/STAT3signaling pathway to regulate autophagy and improve diabetes.Methods 8-week-old SD male rats,fed with high-fat diet for 4 weeks,combined with a single intraperitoneal injection of streptozotocin 40 mg·kg^(-1)to induce an animal model of type 2 diabetes.The Chinese medicine group was given low and high doses of Shenrong Dihuang Decoction,and the western medicine group was given metformin hydrochloride.Observe the general state,body weight,and water consumption of the rats.SD rats in each group were gavage for 4 weeks and then killed by anesthesia.The fasting blood glucose,glycosylated serum protein,triglycerides,cholesterol,and high-density lipoprotein of each group were detected.Cholesterol,low-density lipoprotein cholesterol,free fat,fasting serum insulin.HE staining was used to observe the pathological changes in liver tissues of SD rats.Western Blot method was used to detect the expression of JAK2,STAT3,Beclin-1,LC3,GLUT4 protein.Results Shenrong Dihuang Decoction can improve the general state、weight loss and glucose and lipid metabolism of type 2 diabetic SD rats,meanwhile,it can increase JAK2、STAT3 and GLUT4 protein expression levels,on the other hand,Beclin-1,LC3II/I protein expression levels were reduced.Conclusion The treatment of type 2 diabetes by Shenrong Dihuang Decoction may be achieved by intervening the JAK2/STAT3 signaling pathway,mediating autophagy,increasing the expression level of GLUT4,and improving insulin resistance.
