摘 要:目的 探讨黄芩对复发性流产(Recurrent spontaneous abortion,RSA)模型孕鼠的安胎作用,并阐明其可能的作用机制。方法 分组:空白组、RSA模型组、黄芩治疗组。SPF级CBA/J雌鼠,按照1∶2的比例,随机分为两组:正常妊娠雌鼠组、预RSA模型雌鼠组;然后将BALB/C雄鼠与正常妊娠雌鼠组合笼,交配成功后得到的正常妊娠模型称为空白组;将DBA/2雄鼠与预RSA模型雌鼠组合笼,交配成功后得到复发性流产模型,复发性流产模型孕鼠再随机分成RSA模型组和黄芩治疗组。空白组和RSA模型组给予生理盐水,黄芩治疗组给予黄芩水煎剂,持续给药2周,1次/天。2周后,处死孕鼠,统计胚胎丢失率;HE染色,观察空白组、RSA模型组、黄芩治疗组的孕鼠胎盘组织显微结构的变化;透射电镜(Transmission Electron Microscopy,TEM)观察空白组、RSA模型组、黄芩治疗组的孕鼠胎盘组织超微结构的改变;免疫荧光技术,观察空白组、RSA模型组、黄芩治疗组的诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)、白细胞介素(IL)-6、肿瘤坏死因子-α(TNF-α)、精蛋白-1(Arg-1)、白细胞介素(IL)-10表达。结果 与空白组相比,RSA模型组的胚胎丢失率明显上升(P <0.05);与RSA模型组相比,黄芩治疗组胚胎丢失率明显降低(P <0.05);光镜下观察HE染色,空白组的胎盘在各层次的结构组织均正常;RSA模型组可见蜕膜带部分脱落,细胞坏死,炎性细胞浸润等明显病理改变;黄芩治疗组较RSA模型组的病理改变显著减轻。在透射电镜下观察,空白组胎盘的超微结构在各方面均正常;RSA模型组胎盘的超微结构明显异常,可见不同程度细胞坏死、线粒体肿胀、粗面内质网扩张等;黄芩治疗组胎盘的超微结构未见明显异常。免疫荧光结果显示,与空白组相比,RSA模型组iNOS、IL-6、TNF-α表达明显增高(P <0.05),Arg-1、IL-10表达明显降低(P <0.05);与RSA模型组相比,黄芩治疗组iNOS、IL-6、TObjective To observe the effect of Scutellaria baicalensis on fetal protection in mice with Recurrent Spontaneous Abortion,and to elucidate its possible mechanism.Methods SPF CBA/J females mice were randomly divided into normal pregnant females and RSA model females in a ratio of 1∶2.Normal pregnant females mice mated with BALB/C males mice and the normal pregnancy model established was called blank group.Female RSA mice were mated with male DBA/2 mice to establish recurrent abortion model.The pregnant mice in recurrent abortion model were then randomly divided into RSA model group and Scutellaria baicalensis treatment group.The blank group and RSA model group are given normal saline,and Scutellaria baicalensis treatment group is given baicalensis decoction once per day for consecutive 2 weeks.After 2 weeks,the researcher dissects the mice to observe the number of embryos in the uterus and calculate the rate of embryo loss.The HE staining sections were used to observe the changes of placental tissue microstructure in each group;the Transmission Electron Microscopy(TEM) was used to observe the ultrastructure changes;and the Immunofluorescence Technique was applied to observe the contents of iNOS,IL-6,TNF-α,Arg-1,IL-10.Results Compared with the blank group,the embryo absorption mice of RSA model group increased(P < 0.05);Compared with RSA model group,embryo absorption mice of Scutellaria baicalensis treatment group decreased(P <0.05);HE staining showed that the placenta structure is normal in the blank group;compared with the blank group,the RSA model group showed some obvious pathologicalantifetal changes such as partial shedding of the decidua zone,the necrosis and reduction of giant cell trophoblasts,local necrosis of the sponge trophoblast,and inflammatory cell infiltration;compared with the RSA model group,the degree of pathological changes in the Scutellaria baicalensis treatment group was significantly reduced.Observed under TEM,the placental tissue ultrastructure of the blank group is normal;compared wit
