摘  要:目的探讨辛温通阳中药葱白提取物对高脂血症大鼠腺苷5’-单磷酸激活的蛋白激酶/雷帕霉素的哺乳动物靶标(Adenosine 5’-monophosphate-activated protein kinase/mammalian target of rapamycin,AMPK/mTOR)信号通路表达的影响。方法将30只健康雄性SD大鼠随机分为空白对照组、模型组、葱白低剂量组、葱白中剂量组、葱白高剂量组与辛伐他丁组,除对照组外,其他组均通过脂肪乳灌胃法构建高脂血症模型。对照组与高脂血症模型组每天灌喂生理盐水,葱白低、中、高剂量组分别按照25 mg·kg^(-1)、50 mg·kg^(-1)、70 mg·kg^(-1)灌喂葱白提取物,辛伐他丁组灌喂100 mg·kg^(-1)辛伐他丁;造模成功后生化检测血清中甘油三酯(Triglyceride,TG)、胆固醇(Total cholesterol,TC)、低密度脂蛋白胆固醇(Low-density lipoprotein,LDL-C)、高密度脂蛋白胆固醇(High-density lipoprotein,HDL-C);采用生化方法检测肝脏组织中TC、TG超氧化物歧化酶(Superoxide dismutase,SOD)、琥珀酸脱氢酶(Succinate dehydrogenase,SDH)、及丙二醛(Malondialdehyde,MDA)含量;采用苏木精-伊红染色法(Hematoxylin-eosinstaining,HE)与油红O染色法对肝组织进行病理切片观察;Western blot法检测肝脏组织中AMPK/mTOR信号通路相关蛋白的表达。结果连续给药4周后,与模型组相比,葱白低、中、高剂量组与辛伐他丁组均能降低TC、TG、LDL-C,MDA水平,升高HDL-C,SDH,SOD水平,其中,葱白高剂量组的TC、TG、LDL-C、MDA水平显著降低,而HDL-C、SDH、SOD水平则显著上升(P<0.05)。HE染色与油红“O”染色结果显示,模型组大鼠肝脏细胞脂质严重蓄积,高度脂肪变性,葱白高剂量组在不同程度上改善脂肪蓄积及变性情况。Western blot结果显示,与模型组相比,葱白低、中、高剂量组与辛伐他丁组均能降低mTOR蛋白磷酸化水平,升高AMPK蛋白与ACC蛋白磷酸化水平。结论中药葱白提取物对高脂血症大鼠有明显的降血脂作用,能通过影�Objective To investigate the effect of fistular onion stalker extract on the AMPK/mTOR signaling pathway of hyperlipidemia models.Methods Thirty healthy male SD rats were randomly divided into control group,model group,lowdose extract group,medium-dose extract group,high-dose extract group and Simvastatin group.Rats in all groups except the control group were gavaged with fat milk to construct HLP models.Control group and model group were gavaged with saline everyday.The low-,medium-,and high-dose extract groups were gavaged with the fistular onion stalker extract at 25 mg·kg^(-1),50 mg·kg^(-1),and 70 mg·kg^(-1),respectively.Simvastatin group were gavaged with 100 mg·kg^(-1)Simvastatin.After constructing the HLP models,the serum of each rats on the group were used to detect TC,TG,HDL-C,LDL-C by biochemical examination.The livers of each rats on the group were used to detect the TC,TG,SDH,SOD and MDA by biochemical examination.Stained livers sections with hematoxylin and eosin and oil red O.Western blotting was used to detect the expression of AMPK/mTOR pathway related proteins in liver.Results After continuous administration for 4 weeks,compared with the model group,the low-,medium-,and high-dose extract groups and Simvastatin group can significantly reduce the levels of TC、TG、LDL-C,MDA in serum and rise the level of HDL-C,SDH,SOD(P<0.05).The results of HE and oil red"O"staining showed that the model rats were severe lipid accumulation and a high degree of steatosis in hepatocytes.High-dose extract group improved fat accumulation and degeneration to various degrees.The Western blot assay shows that compared with the model group,in the low-,medium-,and high-dose extract groups and Simvastatin group,the phosphorylation of AMPK protein and ACC protein was significantly increased in rats liver,and the phosphorylation of mTOR protein was significantly reduced.Conclusion The fistular onion stalker extract showed potent hypolipidemic effects on hyperlipidemic rats,which promotes lipid metabolism by affecting the
